Systemic responses induced by Aphanomyces euteiches inoculation in non-nodulated Medicago truncatula roots

ABSTRACT: We performed a split-root experiment with non-nodulated roots of Medicago truncatula (A17 accession) to identify systemic molecular responses induced by the inoculation of the oomycete Aphanomyces euteiches. The root systems of nodulated symbiotic plants were divided into two compartments called the \"local root compartment\" (treated) and the \"systemic root compartment\", from which samples were obtained for RNAseq analysis. Roots, localised in the \"systemic compartment\", were harvested in inoculated or control (mock-inoculated) plants at the same time-points, after 6 hours, 1-, 3- and 6-days after A. euteiches inoculation.

Project description:affy_aphanomyces_medicago - NFP is one of the putative Nod factor receptor and plays therefore a key role in the establishment of symbiosis. nfp allelic mutants are more susceptible to Aphanomyces euteiches (Ae), a root pathogen, than the A17 wild type (WT) line. In this study we want to compare the early plant responses, 1 day post inoculation (dpi) between WT and the nfp2 mutant, in order to identify NFP-dependent gene networks during infection.-Fifteen-day-old A17 or nfp-2 plants were grown in vitro on M medium. The entire roots were then inoculated (or not = controls) with a A. euteiches zoospores (105/ml) and harvested one day after inoculation. Three independent repeats were performed. Project description:affy_infection_medicago - affy_infection_medicago - HAP2a is a transcription factor involved in the regulation of nitrogen fixing nodule organogenesis. hap2a-1 mutant has been shown to be more resistant to Aphanomyces euteiches (Ae), a root pathogen, than the A17 wild type (WT) line. In this study we want to compare both WT and mutant plant responses, 1 and 6 days post inoculation (dpi) between WT and the hap2a-1 mutant, in order to identify HAP2a-dependent gene networks during infection. Data about non inoculated A17 control plants and infected A17 at 1 dpi have been already obtained in another experiment in which the conditions of in vitro culture of M. truncatula plants and ofA.euteiches(Ae) inoculation were exactly the same than the ones used in these experiments.-Fifteen-day-old A17 or hap2a-1 plants were grown in vitro on M medium. Project description:affy_med_2011_10 - affy_med_2011_10 - LYR3 encodes a lysin motif-receptor-like kinase (LysM-RLK) that consists of an extracellular ligand-binding domain composed of three LysM domains, a single transmembrane domain and a cytoplasmic serine/threonine kinase domain. A reverse genetic approach was undertaken to study the role of this gene in the pathosystem Medicago truncatula M-bM-^@M-^S Aphanomyces euteiches (Ae). Results suggest that lyr3-1 and lyr3-2 mutants are more susceptible to the root pathogen Aphanomyces euteiches than the A17 wild type (WT) line. Project description:The experiment aims to identify the response of the mature nodules and denodulated roots of Medicago truncatula /Sinorhizobium medicae md4 to the systemic signaling of plant water deficit. The root systems of nodulated symbiotic plants were divided into two compartments. The N source of N2 fixing plants was exclusively the air. To apply the water stress, we treated half root system with PEG while the other was kept on control conditions. To characterize the systemic responses of the transcriptome to the PEG treatment , nodules and roots of the non-treated half root system were harvested and total RNA isolated for RNAseq analysis. We performed a time-course (6 hours, 1-, 3- and 5-days treatment) .

To characterize the systemic responses of the transcriptome to plant N deficit , nodules and roots of the non-treated half root system were harvested and total RNA isolated for RNAseq analysis. We performed a time-course (6 hours, 1-, 3- and 5-days treatment).

We performed a time-course (1 and 3 days treatment) split-root experiment to identify plant systemic responses: the root systems of nodulated symbiotic plants were divided into two compartments, one of them treated and the other kept on control conditions. Nodules were isolated from the non-treated side of the split root and total RNA isolated for RNAseq analysis.

Time-course transcriptome analysis in roots of N-limited Medicago truncatula plants grown in split-root inoculated with Sinorhizobium medicae

Project description:The experiment aims to analyze the effect of inoculation with Sinorhizobium medicae MD4 of on the transcriptome of the Medicago truncatula roots of N-limited plants. We divided the roots systems into two parts; one of them was supplied with nutrient solution with mineral N (0.5 mM for N-limited plants) whereas the non-N treated part was kept in nutrient solution without mineral N. All roots were inoculated 48h after initiation of the N treatments. We collected the roots the non-N treated side of the split roots system and we isolated total RNA for RNAseq analysis. Root transcriptomes were analyze before inoculation, at 1 and 2 days post-inoculation.
