RNA-Seq of native and FusionRed-transfected canine tumor cell lines after 3 hours C-CPE treatment

ABSTRACT: Claudin (CLDN) proteins are commonly expressed in cancers and accordingly targeted in novel therapeutic approaches. C-terminus of Clostridium perfringens enterotoxin (C-CPE) binds efficiently several claudins and can be used to specifically target cancer cells. This experiment used RNA-seq to profile gene expression changes of native and FusionRed transfected canine tumor cell lines;TihoDTCC0840 (0840) and TihoDProAdCarc0846 (0846) after a 3 hours of C-CPE treatment (20µg/ml).

Gene expression of the mouse choroid plexus epithelium (CPE)

Project description:We investigated the gene expression of the mouse CPE. We isolated CPE cells from three mouse brains with laser dissection microscopy. Next, we performed RNA isolation, amplification, labeling and hybridization against 44k Agilent microarrays. We performed the microarrays against a common reference sample, which was mouse RPE/choroid RNA. We performed 3 mouse CPE replicates from 3 different mouse from the same strain, namely C57BL/6 .

Project description:A microarray analysis was performed to compare the global gene expression profile between C-CPE treated- and untreated- SKOV-3 ovarian cancer cells. SKOV-3 cells were treated with or without C-CPE for 72 hours, and total RNA was extracted and microarray was perfomed to compare the gene profiling changes between C-CPE treated- and untreated- cells. The experiment was performed in triplicate.

Project description:We model processes of wound healing and tumor growth, by studying the effects of normal and cancer epithelial cells on normal fibroblasts and their reciprocal effect on normal keratinocytes in vitro. We find strong parallels between the two processes and compare our observations with transcriptional analysis of 24 clinical samples of squamous cell carcinoma. This dataset contains the tissue culture samples, for the clinical samples, see E-MTAB-1065. Normal human fibroblasts (HF) were cocultured with either normal HK or transformed epithelial cells (HaCaT and FaDu) and their expression profile was compared to the expression profile of HF cultured alone.
