RNAseq of wild type Col-0 Arabidopsis plants and piezo/feronia/camta3 mutants before and 22 minutes after touch treatment

ABSTRACT: RNAseq of wild type Col-0 Arabidopsis plants and piezo/feronia/camta3 mutants before and 22 minutes after touch treatment with a gentle paint brush. The mutants were defective in genes involved in mechanosensing and downstream signalling. Their contribution to touch-responsive signalling was assessed by transcriptome analysis.

Project description:The general stress response is a major transcriptional reprogramming in elicitor response. CAMTA3 is a key regulator of the general stress response, and is guarded by Dominant Suppressor of CAMTA3 1 and DSC2. Here we assay transcriptional response to elf18, an elicitor, at 10 (primary response), 30 (major GSR) and 90 (beginning to be immunity-specific responses) minutes, in wt and in a line mutated in camta3/dsc1/dsc2, thus losing the GSR with only partial autoimmune activation.

A MYC2, MYC3, MYC4-dependent transcription factor network regulates touch responses in plants

Project description:Plants are continuously exposed to environmental triggers, including mechanical stimulation. Our results demonstrate that jasmonic acid (JA)-signalling plays a key role in very early gene expression changes, well before it leads to touch-induced developmental changes. We show that the JA-activated transcription factors MYC2/MYC3/MYC4 co-regulate touch-induced gene expression of 266 genes, many of which peak in induction around 25 minutes and then rapidly decline by 40-60 minutes. ChIP-seq shows that MYC2 dynamically binds hundreds of touch-induced promoters within 25 minutes. Promoter activation assays confirm that MYC2 directly activates these touch-induced promoters. By combining multi-omic data, we have identified a core MYC2/3/4-dependent ‘touch regulon’, containing many previously-unknown MYC2 targets like bHLH19 and ERF109. We show bHLH19 can in turn directly activate the ORA47 promoter, indicating that MYC2/3/4 initiate a hierarchical network of downstream transcription factors. Through hormone profiling we reveal the rapid touch-induced accumulation of JA/JA-isoleucine is directly controlled by MYC2/3/4 in a positive amplification loop regulating JA-biosynthesis genes.

Expression analysis of Acacia koa in response to mechanical stimuli

Project description:The genes induced by mechanical stimuli may be also involved in disease resistance and wood formation and development in Acacia koa. If so, mechanically stressed A. koa may be used as a model to study disease resistance and wood formation and development. Microarray analysis was performed to determine expression levels of 4,000 genes related to disease resistance and wood development in Acacia koa in response to mechanical stimuli (touch). RNA was extracted from two groups of A. koa seedlings, (1) mechanically stressed and (2) unstressed koa seedlings. Each group had two biological replicates (n=2), where n represents pools of approcimately 20 individuals.

Project description:In order to identify novel stress-induced signalling peptides, we searched for Arabidopsis thaliana transcripts encoding short proteins (<150 amino acids) with a predicted signal peptide, which were induced upon biotic elicitor treatment (Bjornson et al., 2021). Through this analysis, we identified an uncharacterised family of peptides with 5 predicted members, which we named CTNIP1 to 5 (pronounced catnip) based on relatively conserved residues within the peptides. CTNIP4 is perceived by Arabidopsis, inducing some hallmark outputs of defence (Pattern-Triggered Immunity) signalling. Mass spectrometric analyses indicated that HSL3 may be the receptor mediating CTNIP recognition. Through assaying transcriptomic responses in wild type and hsl3-1 mutants, we were able to confirm similarity of CTNIP and defence responses at the transcriptional level, and the dependence of these responses on HSL3.
