Single cell RNA-seq of Gif lineage cells in homeostasis and upon injury

ABSTRACT: The purpose of this experiment is to assess and characterize injury response of the gastric chief cells in trancriptomic level. Gif lineage cells (TdTom- GFP+) from Gif-rtTA; TetO-Cre; Rosa-mTmG mice line after Dox administration were isolated from the mouse gastric corpus without injury or at 2d after DMP777-mediated acute injury.

Single cell RNA-seq of Stmn1+ isthmal cells and Pgc+ chief cells to compare and characterize distinct gastric corpus stem cells at molecular level

Project description:The purpose of this single cell experiment is to compare and characterize at molecular level actively cycling stem cells in the isthmus and quiescent stem cells in the base of the mouse stomach corpus. The lineage tracing data from Stmn1-CreERT2 shows that Stmn1+ cells in the corpus isthmus include fast dividing isthmus stem cells with long-term potency. On the other hand, chief cells including Lgr5+ subpopulation can play as quiescent stem cells in the base that are largely quiescent in homeostasis, but are activated upon injury. The isthmus stem cells and chief cells are isolated by Stmn1 and Pgc, respectively, and were subject to single cell RNA-seq experiment.

Transcription profiling by array of homozygous-bred gastric intrinsic factor knockout mice before and after Citrobacter rodententium infection

Project description:Knockout mice were generated that harbour a mutation in the gene encoding gastric intrinsic factor (GIF) a transporter protein necessary for the absorption of vitamin B12/cobalamin (Cbl) from the intestine. The levels of Cbl in the blood of these gif-/- mice exibited a maternal influence dependent on whether they were bred from heterozygous (high Cbl) or homozygous (low Cbl) mothers and this correlated with their susceptibility to infection challenge with Citrobacter rodententium (C.rodentium). Microarrray analysis was conducted on total messenger RNA (mRNA) extracted fron the homzygous-bred gif-/- and wild-type mice with the aim to identify any abnormal genomic expression patterns.

The former formed CD133-positive and CD133-negative cells while the latter only CD133-negative cells, suggesting that CD133-positive cells are mother cells. The former produced differentiated type tumors while the latter undifferentiated types in vivo, indicating a relationship between CD133-expression and glandular structure formation. Gene expression profiling of neural stem cells and their neuronal progeny reveals IGF2 as a regulator of adult hippocampal neurogenesis

Project description:Neural stem cells (NSCs) generate new neurons throughout life in two distinct areas of the mammalian brain: the subventricular zone (SVZ) lining the lateral ventricles and the hippocampal dentate gyrus (DG). How gene expression signatures differ among NSCs and immature neurons within and between these adult neurogenic regions is unknown. We isolated NSCs and their progeny using transgenic mice expressing GFP under the control of the Sox2 promoter (labeling NSCs) and transgenic mice expressing DsRed under the control of the doublecortin (Dcx) promoter (labeling immature neurons). Comparison of the transcriptomes of SOX2+ cells derived from both neurogenic areas revealed that NSCs are highly similar but that functionally significant differences in gene expression exist: IGF2, which is expressed only in SOX2+ cells in the DG but not in the SVZ, is required for proliferation of DG-derived but not SVZ-derived NSCs. Gene expression profiles strongly diverged in immature neurons, and we provide evidence that ephrinB3, which was up-regulated only in the DG but not in the SVZ during neuronal differentiation, regulates the survival of newborn granule cells. Thus, the data provided here show that stem cell populations in the adult DG and SVZ are similar but have unique properties that manifest themselves later during neural differentiation, resulting in distinct neuronal populations Hippocampi and SVZ from 6 week old DCX-DsRed and Sox2-GFP Reporter mice were dissected and cell sorted using FACS. cDNA were generated and analysed using Agilent Platform.
