Microarray of liver tissue from C57BL/6 mouse liver treated with ConA against untreated controls

ABSTRACT: It will be conducive to early diagnosis, appropriate treatment, and improved prognosis for AIH by a comprehensive understanding of the lncRNAs disorders. So, the characterization of these differently expressed lncRNAs, as well as the expression profiling of differently expressed of messenger RNAs, in the AIH mouse model induced by Con A with microarray chip were gained by microarray assay. Con A (15mg/kg, dissolved in pyrogen-free saline) was administrated to the mice in model group (n=4) via tail vein injection. The sham group mice (n=4) were given pyrogen-free saline alone with the same method. All mice were anesthetized lethally at 8h after the administration of Con A or pyrogen-free saline. Hepatic tissues of each mouse were harvested by surgery under low temperature and sterile conditions, and homogenized immediately for extracting total RNA with Trizol Reagent.

Project description:Eight-week-old male pathogen-free BALB/c mice were used for the induction of concanavalin A (Con A)-induced hepatitis. Animals were acclimated for at least 6-7 days to a 12-hour light/dark cycle in a humidity- and temperature-controlled, specific-pathogen-free environment in autoclaved cages. Mice were randomly assigned to two groups (4 in each group) and received Con A type IV (or saline) intravenously via the tail vein at the dose of 20 mg/kg for 2 hours. The animals were sacrificed for blood and liver collection. Blood was taken for transaminase and cytokine measurements. Livers were excised immediately after sacrifice. Part of the livers were immediately snap-frozen in liquid nitrogen and kept at -80Â°C for total RNA isolation and microRNA microarray analysis. miR-674-5p and other miRNAs were experimentally validated to be dysregulated by Con A in mouse livers. Liver samples from four mice treated with Con A and four mice treated with saline were used to acquire the miRNA expression profiling.

Microarray analysis of WT and Batf3-/- CD8alpha dendritic cells from C57BL/6 spleen

Project description:Batf3 regulates key CD8alpha DC-specific genes. WT and KO cells were isolated from pooled spleens of C57BL/6 genetic background mice, that were either untreated, or injected with recombinant murine IL-12 in pyrogen-free saline at 2.5 ug/mL. Mice were injected i.p. with 0.5 ug of IL-12 once and sorted after 3 days. CD8alpha+ dendritic cells were sorted as B220- NK1.1- CD172a- CD11c+ MHCII+ CD24+ DEC205+ CD8alpha+ cells.

Project description:Translational profiling of mouse cardiac tissue treated with 15mg/kg doxorubicin in 10 ml/kg saline over an acute time course (0.5-120 hours) compared to time matched control animals treated with 10ml/kg saline. Before microarray analysis, RNA is separated on a sucrose density gradient into those mRNAs activity undergoing translation (polysomes) and those not (monosomes) with control monosomes and treated monosomes on one set of arrays, and polysome control and polysome treated on another set of microarrays. Thealized Log2 of the monosomes was subtracted from the respective Log2 of the polysomes (on Series record). Time points studied were 0.5, 1, 2, 12, 24 and 120 hours following dosing, biological replicates n=3 independent animals at each time point, technical replicates (reverse labelling) n<1. One array printed onto two slides (A and B), one replicate per array.

Proteomic results of Lactobacillus plantarum P8 alleviating dexamethasone-induced oxidative stress in broiler liver

Project description:The broilers were randomly allotted to four treatment groups (Con, DEX, P8, and DEX+P8 groups) with 10 replicates per group (10 broilers per replicate). Broilers in the Con and DEX groups were fed a basal diet. Broilers in the P8 and DEX+P8 groups were fed a basal diet containing 1 × 108 CFU/g P8. At 16 days of age, broilers in the DEX and DEX+P8 groups were injected with 3 mg/kg body weight DEX (200 μL), whereas broilers in the Con group were injected with an equal volume of saline.

Azoxymethane-induced colorectal tumorigenesis in hypertensive rat models

Project description:To compare the tumor susceptibility between the hypertensive Dahl salt-sensitive rats (S) rat and S.LEW(10)x12x2x3x5 (S.LEW) congenic strain, one of the S.LEW congenic strains our laboratory previously constructed on rat chromosome 10 [Hypertension. 2007 Nov;50(5):891-8], Azoxymethane (AOM)-induced colon tumorigenesis in rats was assessed. At six weeks of age, both S and S.LEW congenic rats received intraperitoneal (IP) injection of AOM in sterial saline at a dose of 15mg/kg body weight once a week for two consecutive weeks. At 30 weeks of age, all the rats were euthanized through carbon dioxide inhalation. The colon tissue was harvested, dissected longitudinally, and washed with saline. The number and size of the colon tumors were recorded. We found that the total number of colon tumors was significantly higher in the S.LEW congenic strain compared with the S rat. To further study the genetic and molecular mechanisms causing the higher tumor susceptibility in the S.LEW congenic strain, we have employed the whole genome microarray expression profiling to identify genes and signaling pathways that are differential between the S and S.LEW congenic strain. At six weeks of age, both S and S.LEW congenic rats received intraperitoneal (IP) injection of AOM in serial saline at a dose of 15mg/kg body weight once a week for two consecutive weeks. At 30 weeks of age, all the rats were euthanized through carbon dioxide inhalation. The colon tissue was harvested, dissected longitudinally, and washed with saline. RNA from the harvested colons of 3 S and 3 S.LEW congenic rats were prepared for the microarray study.

Amydala transcriptome changes in germ-free mice

Project description:We sequenced mRNA from 12 samples extracted from mouse amygdala tissue to generate the first amygdala-specific murine transcriptome for germ-free mice (GF), conventionally raised controls (CON) and germ-free mice that have been colonized with normal microbiota from postnatal day 21 (exGF). Equal amounts of RNA from two to three animals were pooled to yield 4 samples per group (CON, GF, and exGF). Pairwise comparisons for CONvsGF, CONvsexGF, GFvsexGF were performed using DESeq2.
