RNA-seq of ex vivo cultured lung CD140a+ mesenchymal cells (fibroblasts) upon stimulation by IL-1β

ABSTRACT: To gain a better understanding of the role of Interleukin-1β (IL-1β) in lung CD140a+ mesenchymal cells (fibroblasts) modulation, we performed RNA-seq to compare the transcriptomes of IL-1β-treated and control lung CD140a+ mesenchymal cells (fibroblasts).

CD140a+ human glial progenitor cells

Project description:Glial progenitor cells (GPCs) pervade the human brain. These cells express gangliosides recognized by MAb A2B5, and some but not all can generate oligodendrocytes. Since some A2B5+ GPCs express PDGFa receptor (PDGFRa), which is critical to oligodendrocyte development, we asked if PDGFRa-directed sorting might isolate oligodendrocyte-competent progenitors. We used FACS to sort PDGFRa+ cells from the second trimester fetal human forebrain, based on expression of the PDGFRa epitope CD140a. CD140a+ cells could be maintained as mitotic progenitors that could be instructed to either oligodendrocyte or astrocyte phenotype. Transplanted CD140a+ cells robustly myelinated the hypomyelinated shiverer mouse brain. Microarray confirmed that CD140a+ cells differentially expressed PDGFRA, NG2, OLIG1/2, NKX2.2 and SOX2. Some expressed CD9, thereby defining a CD140a+/CD9+ fraction of oligodendrocyte-biased progenitors. CD140a+ cells differentially expressed genes of the PTN-PTPRZ1, wnt, notch and BMP pathways, suggesting the interaction of self-renewal and fate-restricting pathways in these cells, while identifying targets for their mobilization and instruction. 10 samples, 5 CD140a+, and 5 CD140a- sorted samples for individual fetal human brain

Transcription profiling of mouse alveolar type II epithelial cells

Project description:The importance of alveolar type II epithelial cells (AEC II) in all aspects of the induction and regulation of respiratory immune responses has become increasing appreciated. We determined the genome-wide profile of gene expression in response to immune mediated airway inflammation. Overall, our results provide detailed description of specific AEC II gene expression under disease conditions. Differentially expressed genes of diverse molecular functions have been identified that may be critical for numerous physiologic activities, some of which may be currently unappreciated. Data obtained by such analysis will help to understand the function of these important immune cells in the respiratory system and may point out strategies for intervention in the progression of inflammatory diseases. We used microarrays to detail changes in the gene expression profile in alveolar type II epithelial cells during interstitial airway inflammation and identified a wide variety of differentially expressed genes during this process. Experiment Overall Design: Alveolar type II epithelial cells from the lung of healthy SPC-HA and diseased SPC-HAxTCR-HA mice were isolated by enzymatic tissue desintegration followed by one step negative selection using a MoFlow cell sorter. RNA was extracted and hybridization on Affymetrix microarrays was performed.

Project description:Comparison of DamID profiles and LAD patterning across cell types reveals regions of variable LADs LmnB1 interactions at the nuclear periphery of C57Bl/6 pro-B cells and fibroblasts were identified by DamID in duplicate for both pro-B and fibroblasts (4 samples total). For each sample, Dam-LmnB1/Dam-only Log2 ratios of chromosomes 11 and 12 were calculated, lifted over to mm9, duplicates were normalized and averaged together.

Project description:The purpose of this experiment was to determine the transcriptional differences between neural progenitor cells from neonatal lung injury mice vs. control mice, as well as neonatal lung injury mice treated with umbilical-cord mesenchymal stromal cell extracellular vesicles vs. neonatal lung injury mice treated with placebo (PBS). Neural progenitor cells were isolated from the subventricular zone and hippocampus and cultured for 2 consecutive neurosphere assays. RNA was then extracted from the cells, and the microarray labelling, hybridization, and scanning was conducted by the Génome Québec Innovation Centre (Montréal, Canada).

Proteomic characterization of mesenchymal lung cells

Project description:Mesenchymal cells in human lung are heterogneous and can display a large variation in both functional properties and spatial localization. However it remain unclear to witch extent these observed cell population represent different cell types or just different cellular states. Furthermore, the lack of specific markers has hampered the progress within the field. In this study we aimed to investigate the proteome of mesenchymal cells derived from human lung as a starting point to learn more about mesenchymal subsets and identify novel markers.
