RNA-Seq of the liver of mice after 3-week treadmill training versus sedentary control

ABSTRACT: This study investigates the effects of exercise and genetic predisposition on the transcriptomic profile of the liver of long-term selected marathon (DUhTP) and non-inbred (DUC) mice. In the exercise group, mice underwent treadmill training for three weeks. For the sedentary control group, mice were kept under minimal physical activities. For the 3-week training program, the mice were running five days per week (Monday to Friday) starting at age of 49 days after birth (Walz et. al. 2021). All mice used in this experiment were male and sacrificed at day 70 of life for tissue sampling.

Forced intensity-controlled endurance training on a small-animal treadmill machine inducing murine cardiac hypertrophy

Project description:Endurance training is associated with decreased cardiovascular-related morbidity and mortality. Cardiac hypertrophy is an adaptive mechanism of the heart in response to volume and pressure overload. Nonetheless, murine exercise models for cardiac hypertrophy are still under discussion. Using a small-animal treadmill, a forced intensity controlled training model was conducted to characterize cardiac hypertrophy in mice utilizing multimodal analyses. Wild-type male C57BL/6 mice age between 5-8 weeks old were subjected to 8 weeks of forced endurance training using a small-animal treadmill or sedentary age-matched control. Transcriptomic analysis of 8-week training hearts was evaluated using RNA sequencing. RNA sequencing revealed several genes involved in the cell cycle, apoptosis, contractile protein expression, and organ growth that were among the most differentially regulated genes after 8-weeks of exercise.

Next Generation Sequencing Transcriptomic Analysis (RNA-seq) of Thoroughbred horse at rest, after exercise and after training

Project description:A single bout of exercise induces changes in gene expression in skeletal muscle. Regular exercise results in an adaptive response involving changes in muscle architecture and biochemistry, and is an effective way to manage and prevent common human diseases such as obesity, cardiovascular disorders and type II diabetes. Our study is a transcriptome-wide analysis of skeletal muscle tissue in a large cohort of untrained Thoroughbred horses before and after a bout of high-intensity exercise and again after an extended period of training. We hypothesized that regular high-intensity exercise training primes the transcriptome for the demands of high-intensity exercise.

Identification of exercise-related genes in mice exposed to cigarette smoke

Project description:Cigarette smoke (CS) is the major risk factor for COPD and is linked to cardiopulmonary dysfunction. Exercise training, as part of pulmonary rehabilitation, is recommended for all COPD patients. It has several physiological benefits, but the involved mechanisms remain poorly defined. Here, we employed transcriptomic profiling and examined lung endothelium to investigate novel interactions between exercise and CS on cardiopulmonary alterations. Mice were exposed to 20 weeks of CS, CS + 6 weeks of high-intensity interval training on a treadmill or control. Lung and cardiac (left and right ventricle) tissue were harvested, and RNA-sequencing was performed and validated with RT-qPCR. Immunohistochemistry assessed pulmonary arteriolar changes. Transcriptome analysis between groups revealed 37 significantly regulated genes in the lung, 21 genes in the left ventricle, and 43 genes in the right ventricle (likelihood-ratio test). Validated genes that showed an interaction between exercise and CS included angiotensinogen (p=0.002) and resistin-like alpha (p=0.019) in left ventricle, with prostacyclin synthetase different in pulmonary arterioles (p=0.004). Transcriptomic profiling revealed changes in pulmonary and cardiac tissue following exposure to CS, with exercise training exerting rescue effects. Exercise-regulated genes included angiotensinogen and resistin-like alpha. However, it remains unclear if these represent potential candidate genes or biomarkers involved during pulmonary rehabilitation.

An Oleuropein-based olive leaf extract enhances the muscle mitochondrial response to acute moderate – but not maximal – intensity exercise in humans

Project description:Sprint interval exercise (SIE) induces mitochondrial adaptations in skeletal muscle, comparable to or greater than moderate-intensity continuous exercise (MICE), despite lower training volume. These effects are partly due to enhanced mitochondrial bioenergetics, including increased Ca²⁺ uptake and pyruvate dehydrogenase (PDH) activation. Oleuropein (OLE), a compound from olive leaf extract, also promotes mitochondrial Ca²⁺ uptake and PDH activation in mice. The current study tested whether OLE could enhance these adaptations in humans during MICE or SIE. In a double-blind crossover design, healthy males performed either MICE or SIE with OLE supplementation. To elucidate the acute molecular response of skeletal muscle following exercise, we performed mass spectrometry-based proteomic analysis.

Transcription profiling of heart tissue from rats that have undergone treadmill exercise or have been sedentary for different lengths of time

Project description:Seventy two rats were randomized to twelve independent groups of rats (n = 6 in all groups) where half of the rats were chosen for training (treadmill exercise training 1.5 hour 5 days a week, in 1, 4, 24, 48 days and for 4 and 8 weeks) or sedentary (no training) as described in detail by Wisloff et al. (Intensity-controlled treadmill running in rats: VO(2 max) and cardiac hypertrophy. Am J Physiol Heart Circ Physiol. 2001 Mar;280(3):H1301-10.) or Kemi et al.(Aerobic fitness is associated with cardiomyocyte contractile capacity and endothelial function in exercise training and detraining. Circulation. 2004 Jun 15;109(23):2897-904. Epub 2004 Jun 1.). The rats were scarified one hour after training (1, 4, 24 and 48 days) or 24 hours after training (4 and 8 weeks). Biopsies form left ventricle was taken form all rats and stored on -80°C for preparation of RNA. The same procedures were performed for the control/sedentary rats which were sacrificed at the same time as the exercised rats.

Treadmill training regulates bone metabolism and osteogenic differentiation(miRNA Array)

Project description:MicroRNAs (miRNAs) are important in the process of exercise influencing bone metabolism. The present study aimed to detect and select differentially expressed miRNAs in the bone tissues of mice trained on a treadmill, predict the target genes of these differentially expressed miRNAs and lay a foundation for exploring the effect of treadmill training on bone metabolism through miRNAs. In this experiment, after the mice were trained on a treadmill for 8 weeks, the mechanical properties of mouse femur bone were assessed, and the alkaline phosphatase (ALP) activity and osteocalcin (OCN) protein levels of the bone were assayed. miRNA microarray and reverse transcription-quantitative (RT-q)PCR were performed to select and validate differentially expressed miRNAs in the bone, and the target genes of these miRNAs were predicted with bioinformatics methods. In addition, the differentially expressed miRNAs in the bone tissues were compared with those in mechanically strained osteocytes in vitro.
