Establishment of patient-derived bone organoids to recapitulate and dissect Hurler syndrome's skeletal disease

Project description:Mucopolysaccharidosis type I is a disease caused by the mutation of the IDUA gene, encoding the alpha-L-Iduronidase enzyme. Accumulation of glycosamminoglycans (GAGs) due to IDUA deficiency results in a complex skeletal phenotype referred to as dysostosis multiplex. The cellular and molecular pathogenetic mechanisms underlying dysostosis multiplex are still poorly understood. In this study we adopted in vitro and in vivo experimental systems based on patient derived bone marrow stromal cells (BMSC) to generate chondroid and bone rudiments. Transcriptome analysis of chondroid pellet at 5 weeks generated in vitro with BMSCs derived from HD and MPS IH could help to elucidate molecular mechanisms underlying the skeletal phenotype of Hurler Syndrome.

Characterization of arterial dendritic cells in giant cell arteritis and polymyalgia rheumatica

Project description:Giant cell arteritis and polymyalgia rheumatica are two frequently overlap conditions. A pathophysiological continuum between these two diseases has been highlighted by the demonstration of activated dendritic cells in the arterial wall of PMR and GCA patients. However, in PMR, activation of these dendritic cells was not associated with inflammatory infiltration of the arterial wall. The aim of this study was to characterize arterial wall dendritic cells during PMR and GCA. BulK RNA sequencing was performed from FFPE sections of temporal artery biopsies from control (n = 3), PMR (n = 5) and GCA (n = 3) patients. Total RNA was extracted from FFPE tissue sections using the Maxwell RSC RNA FFPE kit (Promega). Ribosomal RNA-depleted RNA was used for library preparation with the NEBNext Ultra II Directional RNA library prep kit for Illumina (New England Biolabs). Libraries were paired-end sequenced (2*76 base pairs) on a NextSeq500 device (Illumina), with a read depth of 40 million. Kallisto software was used for quantifying transcript abundance from RNA-seq data against GRCh38 cDNA reference transcriptome from the Ensembl database, v96.

RNA-seq of Dm-aKG treated intestinal stem cells (ISCs) and controls

Project description:The object of this study was to investigate how aKG impacts mouse intestinal stem cell (ISCs) function and fate decisions in vivo. Mice were treated for 7 days with twice daily aKG IP injections after which ISCs were FACS sorted and transcriptomically anayzled. This revealed a striking set of secretory genes upregulated in stem cells after aKG treatment.

Ablation of Lgr5-positive cells in intestinal polyps from ApcMin;Lgr5DTR mice

Project description:We performed RNAseq on intestinal polyps from diptheria toxin-treated ApcMin;Lgr5DTR mice to investigate the effect of an acute selective pressure on stem cell populations in intestinal lesions. Lgr5+ cells in the ApcMin;Lgr5DTR mice were ablated with a single intraperitoneal dose of diphtheria toxin in saline (50 μg/kg), and samples were collected after 24 hours and after 5 days. Untreated ApcMin mice were used as control. Intestinal polyps were excised and collected for RNA sequencing.
