Transcript Profiling after a Genetically-induced Elevation of Trehalose 6-Phosphate Reveals a Plethora of Direct and Indirect Responses

ABSTRACT: We aim at understanding the Tre6P-specific signalling responses by transiently inducing the expression of the bacterial TPS (otsA) protein at the beginning of the day, leading to an induced rise in Tre6P levels. Treated plants- 2% ethanol spray. non-treated - water spray. empty-vector control - alcR (all previously described in Martins et al. (2013) and in Figueroa et al. (2016).

Using microarrays we studied the crosstalk between low R:FR and SA at the global gene expression level in Arabidopsis thaliana. Plants were exposed for 2 h. to the following treatments: high R/FR with mock spray, low R/FR with mock spray, high R/FR with SA spray, low R/FR with SA spray. Gene expression was determined in petioles.

Global DNA methylation analysis of mouse totipotent potential stem cells, extended pluripotent stem cells and mouse embryonic stem cells

Project description:It is challenging to derive totipotent stem cells in vitro that functionally and molecularly resemble cells from totipotent embryos. Here, we report that a chemical cocktail enables the derivation of totipotent-like stem cells, designated as totipotent potential stem (TPS) cells, from 2-cell mouse embryos and extended pluripotent stem cells that can be stably maintained long-term in vitro. TPS cells shared features with 2-cell mouse embryos in terms of totipotency markers, transcriptome, chromatin accessibility and DNA methylation patterns. In vivo chimeric assays show that these cells have embryonic and extraembryonic developmental potentials at the single cell level. Moreover, TPS cells can be induced into blastocyst-like structures resembling preimplantation mouse blastocysts. Mechanistically, inhibition of HDAC1/2 and DOT1L activity and activation of RARγ signaling are important for inducing and maintaining totipotent features of TPS cells. Our study opens up a path toward fully capturing totipotent stem cells in vitro.

Project description:Inducible overexpression of STM by AlcR / AlcA system. Plants harboring 35S::AlcR/AlcA::GOI (GUS control, STM and STM-VP16)constructs were grown in continous light for 12 days and induced with 1% Ethanol. After 12h of EtOH treatment, seedlings were dissected and RNA was processed from the shoot apex and young leaves. Affymetrix Ath1 arrays were hybridized in duplicates from each experiment.

Project description:In order to evaluate the genome differences and find the more tolerant cultivar, first eleven Malaysian rice cultivars namely, MR219, MR276, MR220, MR211, MR219-4, MR253, Q50, Q74, MR159, Masuri and MR263 were subjected under water deficit. Then, based on the morphological and physiological traits, the more drought-tolerant and -susceptible cultivars were screened and time-course gene expression profiling established by a comprehensive transcriptome database sequencing of the leaf RNA of tolerant rice. The current investigation provides pivotal data for understanding the rice drought tolerance mechanisms.
