CT26 sorted in vivo tumor cells with Trex1 KO

ABSTRACT: The DNA exonuclease TREX1 degrades endogenous cytosolic DNA. Cytosolic DNA triggers the cGAS/STING pathway which increases type I interferon. To investigate the physiological significance of TREX1 loss on in vivo tumor growth, we implanted control and TREX1-deficient CT26 tumor cells into immunocompetent BALB/c hosts.Tumor cells were collected 7 days after tumors reached around 200mm3.

Project description:To mimic menopause hormone therapy and compare the effects of estetrol (E4) to the ones of estradiol (E2), ovariectomized mice were treated for 2 weeks with E2, a physiological dose of E4 (E4low) or a supra physiological dose of E4 (E4hi). Non treated ovariectomized mice were used as control. Epithelial cells and fibroblast cells from uterus and vagina were sorted based on surface markers and RNA was collected in duplicates. RNAseq was performed for each replicate.
