Osteoblast mineralisation

ABSTRACT: Bone mineralization is a carefully orchestrated process, regulated by a number of promoters and inhibitors that function to ensure effective hydroxyapatite formation. Here we sought to identify new regulators of this process through a time series microarray analysis of mineralising primary osteoblast cultures over a 27 day culture period.

Capacitive electrical stimulation effects on osteoblast-released matrix microvesicles

Project description:Passive implants currently used in the clinics cannot prevent failure rates and inherent revision arthroplasties. Novel bioelectronic devices that include biophysical stimulators (such as electric stimulators) and sensing systems are desired since these will allow for long-term monitoring and control of the bone-implant interface, in a personalized manner. We have developed acting-sensing dual systems operated at high frequency (HF) that are able to stimulate osteoconduction in pre-osteoblasts and osteoinduction in human adipose-derived mesenchymal stem cells (hASCs). The project included – to our knowledge – the first-time proteomic analysis of microvesicles secreted from osteoblasts electrically-stimulated in vitro by a capacitive stimulator of thin interdigitated electrodes delivering an electrical 60 kHz HF stimulation, 30 min/day. Results revealed regulation of osteodifferentiation and mineralization-related proteins (e.g. Tgfb3, Ttyh3, Itih1, Aldh1a1).

Transcription profiling of mouse osteoblast cell from wild-type and retinoblastoma tumor suppressor(Rb) knock-out.

Project description:Here we report the characterization of a novel role for the retinoblastoma protein (pRb) as a regulator of osteoblast adhesion. Abrogation of pRb in osteoblasts resulted in aberrant cadherin expression and loss of adherens junctions. This produced defects suggestive of a transformed phenotype such as impaired cell-to-cell adhesion, loss of contact-dependent growth arrest, and the capacity to evade anoikis. This also resulted in profound abnormalities in bone structure. Consistent with this, microarray analyses showed that pRb regulates a wide repertoire of osteoblast cell adhesion genes. In addition, pRb loss also resulted in altered expression and function of several known regulators of cellular adhesion and adherens junction assembly, such as the Rho GTPase Rac1 and the merlin tumor suppressor. Taken together, our results show that pRb controls cell adhesion by regulating the expression and adherens junction components and by regulating the function of molecules involved in adherens junction assembly and stability. Microarray results helped us to portrait the overall influence on cell adhesion via both individual genes and pathway analysis. Experiment Overall Design: MC3T3 cells were obtained from immortalizing primary cultured mouse osteoblast cells by using 3T3 protocol. There are 3 biological replicates for each group, 6 samples in total were analyzed.

Osteoblast Gene Expression in Response to Bicc1 Knockdown

Project description:We recently identified Bicc1 as a regulator of osteoblast differentiation and bone mass. Bicc1 encodes an RNA-binding protein. Here, we have used siRNA to decrease Bicc1 expression in primary calvarial osteoblasts. Illlumina microarrays were then used to profile global gene expression changes. The experiment consisted of 16 total samples in 4 treatment groups (N=4 replicates per treatment). The treatment groups were osteoblasts treated with 1). control scrambled siRNA, 2) Bicc1_1 siRNA, 3) Bicc1_2 siRNA and 3) Bicc1_3 siRNA. Cells were isolated, transfected and 48 hours later RNA was isolated and hybridized to arrays.

GPM6B regulates osteoblast differentiation and induction of mineralization by controlling cytoskeleton and matrix vesicle release.

Project description:Analysis of GPM6B silenced osteogenic hMSC at gene expression level. Analysis is showing significant changes in genes involved in cytoskeleton organization and biogenesis. Immunocytochemistry confirms changed distribution of actin filaments and change in shape and in size of focal adhesions upon GPM6B silencing. Moreover, we demonstrated that production and release of ALP-positive matrix vesicles (MVs) was reduced. In conclusion, we identified GPM6B as a novel regulator of osteoblast differentiation and bone formation and thereby demonstrating the significance of cytoskeleton organization for MV production and eventual mineralization. Total RNA obtained from osteogenic hMSC, 7days after shRNAi lenti viral transductions with 3 different GPM6B silencing constructs (sh1, sh2, sh3) compared to 3 different controls (shC:non-silencing shRNA, tGFP: tGFP control vector and Mock: mock transduced cells)

Next Generation Sequencing Facilitates Quantitative Analysis of Osteoblast Mineralization Transcriptomes

Project description:We report the application of next generation sequencing technology for high-throughput profiling of mRNA, circRNA and lncRNA in hFOB 1.19 cells during the mineralization. Osteoblast RNA profiles in different process of mineralization, which is day 0, day 3, day 6 and day 9, respectively. The transcriptome sequencing on an Illumina HiSeq X Ten platform were carried out at Novogene Bioinformatics Technology Co., Ltd. (Beijing, China). Burrows–Wheeler Aligner (BWA) followed by ANOVA (ANOVA) and TopHat followed by Cufflinks. In the process of osteoblast mineralization, a total of 8464 dysregulated circRNAs were identified, of which 6332 circRNAs were upregulated and 2132 circRNAs were downregulated. In addition, 1633 differentially expressed mRNAs were also identified, with 1027 mRNAs upregulated and 606 mRNAs downregulated. This study provides a framework for the application of NGS profiling towards characterization of osteoblast mineraliztion.

Transcription profiling by array of human trabecular bone osteogenic cells

Project description:The aim of this study was to describe the gene expression patterns related to the differentiation and mineralization of bone-forming cells, including activation and/or repression of osteogenic or non-osteogenic pathways, remodeling of cell architecture, cell adhesion, cell communication, and assembly of extracellular matrix. The study implied patient selection, tissue collection, isolation and culture of human marrow stromal cells (hMSC) and osteoblasts (hOB), and characterization of bone-forming cells. RNA samples were collected at defined time points, in order to understand the regulation of gene expression during the processes of cell differentiation/mineralization that occur during bone repair. Transcriptome analysis was performed by using the Affymetrix GeneChip microarray technology platform and GeneChip Human Genome U133 Plus 2.0 Array. Our results help to design a gene expression profile of bone-forming cells during specific steps of osteogenic differentiation. These findings offer an useful tool to monitor the behaviour of osteogenic precursors cultured in presence of exogenous stimuli, i.e. growth factors, or onto 3D scaffolds for bone engineering. Moreover, they can contribute to identify and clarify the role of new genes for a better understanding of the molecular mechanisms regulating osteogenesis. Experiment Overall Design: Differentiated osteoblasts (hOB) were obtained from trabecular bone fragments of four patients. hOB cultures were maintained in mineralization medium containing beta-glycerophosphate, and collected at different time points. The experimental protocol was specifically devised to mark four steps of hOB mineralization (HM). The reference sample consisted in confluent hOBs before the addition of mineralization medium (HM1).
