Bioengineering of a human innervated cardiac muscle model

Project description:Cardiac autonomic neurons control cardiac contractility. Dysregulation of the autonomic nervous system can lead to sympathetic overdrive resulting in heart failure and an increased incidence of fatal arrhythmias. Here, we introduce innervated engineered human myocardium (iEHM), a novel model of neuro-cardiac junctions, constructed by fusion of a bioengineered neural organoid patterned to autonomic nervous system (SNO) and engineered human myocardium (EHM). Projections of sympathetic neurons into EHM formed presynaptic terminals in close proximity to cardiomyocytes and the extensive vascular network co-developing in the tissues. Contractile responses to optogenetic stimulation of the accordingly engineered neuronal component demonstrated functionality of the engineered neuro-cardiac junctions. Modeling long-QT 2 related to KCNH2 mutations in iEHM revealed early afterdepolarizations and autonomic neuron hyperactivity, suggesting a contribution of the autonomic nervous system to hERG-channel dysfunction-related arrhythmias. Collectively, our data demonstrate the utility of iEHM in the delineation of neuron-cardiomyocyte crosstalk at the tissue level and may serve as an important step towards the engineering a human brain-to-heart axis in a dish.

Single-cell RNA sequencing (scRNA-seq) profiles of fetal human cortical neurons

Project description:The dataset consists of single-cell RNA sequencing (scRNA-seq) profiles of fetal human cortical neurons (10 weeks gestation) under six experimental conditions: Sample 1 – Control: Untreated cells. Sample 2 – PFF treated: Cells exposed to alpha-synuclein pre-formed fibrils (PFF) for 14 days. Sample 3 – PFF + B36D: Cells treated with PFF and the peptide inhibitor B36D. Sample 4 – B36D alone: Cells treated with B36D without PFF. Sample 5 – PFF + S62: Cells treated with PFF and the peptide inhibitor S62. Sample 6 – S62 alone: Cells treated with S62 only. Culture and treatment timeline: All cells were initially cultured for 7 days before treatment. Treatments were applied for 14 days. On day 21 in vitro, cells were dissociated for single-cell RNA sequencing. This dataset captures the gene expression changes associated with PFF-induced pathology and the potential rescue effects of peptide inhibitors B36D and S62 in human cortical neurons. It allows comparisons between control, disease-mimicking, and treatment conditions at single-cell resolution.

New insights into the molecular actions of Grosheimin, Costunolide, α- and β- Cyclocostunolide on primary cilia structure and Hedgehog signaling

Project description:Human retinal pigment epithelial (RPE) cells were treated with the sesquiterpene lactones α-cyclocostunolide (α-C) or Grosheimin to investigate their effects on primary cilium structure and associated signaling pathways. Sesquiterpene lactones (SLs) are plant-derived metabolites known for diverse pharmacological activities. Previous studies have shown that certain SLs can alter primary cilium morphology; however, their impact on ciliary signaling remains unclear. This dataset explores the transcriptional changes induced by α-C and Grosheimin in RPE cells. RNA sequencing was performed to identify gene expression changes related to ciliogenesis, microtubule organization, and Hedgehog (Hh) signaling. These data contribute to understanding how structurally distinct SLs modulate ciliary function and may inform future studies on ciliopathies and tumor biology.

RNA sequencing of non–small cell lung cancer (NSCLC) cell line H1299 following chloroquine (CQ) treatment

Project description:Functional lysosomes are essential for tumor growth. CQ, a widely used lysosome inhibitor, has been combined with various other therapeutics for testing in cancer patients. However, no studies have reported enhanced antitumor effects from these treatments. To investigate how tumor cells survive CQ-mediated lysosome inhibition, we conducted transcriptome analysis using RNA sequencing (RNA-seq) on a NSCLC H1299 cell line to compare gene expression changes between control (vehicle) and CQ treatment groups.

RNA-Seq profiling of human embryonic stem cells carrying different levels of mtDNA heteroplasmy cultured in mTeSR Plus and E8 media.

Project description:Mitochondrial DNA mutations in components of the electron transport chain disrupt mitochondrial activity and can alter how cells take up and utilize nutrients. To investigate how mtND6 (Complex I) mutations reshape the transcriptional profile of human pluripotent stem cells, we performed bulk RNA sequencing on hESCs carrying levels of mtND6 mutations: wild-type, 5%, 56%, and 77% heteroplasmy. All four lines were profiled in mTeSR+, and wild-type and 77% mutant hESCs were additionally profiled in Essential 8 (E8) medium to identify transcriptional changes robust to differences in culture nutrient composition. Each sample was sequenced in biological triplicate (n = 18). Total RNA was poly(A)-selected, and libraries were sequenced on an Illumina NovaSeq platform, paired-end read, 20 million reads per sample.
