3' sequencing following time-course inhibition of ADMA+SDMA or CDK11+TOPK in LU-99 cells

ABSTRACT: Numerous RNA-binding proteins involved in alternative polyadenylation are arginine methylation substrates. Therefore we aimed to study how alternative polyadenylation is impacted by preventing arginine methylation, and how this phenotype develops at various time points. We treated LU-99 cells with 100 nM LLY-283 to inhibit symmetric dimethylation and 3 μM GSK-715 to inhibit asymmetric dimethylation and harvested cells over 24 hour time periods following this inhibition (24, 48, 72 and 96 hours). We also treated cells with 50 nM TOPK and CDK11 inhibitor OTS964 as a negative control and harvested cells after 24 hours incubation. We sequenced the 3' ends of poly-adenylated mRNAs extracted from these cells.
