3' sequencing following 96hrs ADMAi, SDMAi or ADMAi+SDMAi in LU-99 cells

ABSTRACT: Numerous RNA-binding proteins involved in alternative polyadenylation are arginine methylation substrates. There are two forms of dimethylation - asymmetric dimethylation (ADMA) and symmetric dimethylation (SDMA). We aimed to study how alternative polyadenylation is impacted by preventing arginine methylation, and compare which form of dimethylation regulates APA to the largest extent. Therefore, we treated LU-99 cells with 100 nM LLY-283 to inhibit SDMA, 3 μM GSK-715 to inhibit ADMA, or both 100 nM LLY-283 and 3 μM GSK-715 to inhibit both forms of dimethylation, and harvested cells after 96 hours. We sequenced the 3' ends of poly-adenylated mRNAs extracted from these cells.
