3' sequencing following arginine methylation inhibition in a panel of cancer cell lines

ABSTRACT: Numerous RNA-binding proteins involved in alternative polyadenylation are arginine methylation substrates. Therefore we aimed to study how alternative polyadenylation is impacted by preventing arginine methylation, and assess how conserved this phenotype is across a panel of cancer lines with differing genetics and tissue origins. We treated 10 cell lines (GP2D, HCT116 p53 +/+, HCT116 p53 -/-, LU-99, MCF7, NCI-H838,PANC0403, SK-MEL-2, SUM149PT, U2OS) with 100 nM LLY-283 to inhibit symmetric dimethylation and 3 μM GSK-715 to inhibit asymmetric dimethylation, and harvested cells after 72 hours. We sequenced the 3' ends of poly-adenylated mRNAs extracted from these cells.
