RNA-sequencing from human salivary glands from Sjogren's disease patients recruited in the TRACTISS randomized clinical trial

ABSTRACT: Sjogren's disease patients received Rituximab (anti-CD20 depleting monoclonal antibody) or placebo treatment. This study aims to identify the transcriptomic features of salivary gland biopsies, the target tissue of the autoimmune response in Sjögren’s disease. It examines transcriptomic changes in salivary gland tissue from patients treated with Rituximab compared to those in the placebo group, alongside longitudinal changes pre/post treatment.

Aquaporin gene therapy corrects Sjogren's syndrome phenotype in mice

Project description:Primary Sjögren’s syndrome (pSS) is a chronic autoimmune disease that is estimated to affect 35 million people worldwide. Currently, no effective treatments exist for Sjögren’s syndrome, and there is a limited understanding of the physiological mechanisms associated with xerostomia and hyposalivation. The present work revealed that aquaporin 5 expression, a water channel critical for salivary gland fluid secretion, is regulated by bone morphogenetic protein 6. In- creased expression of this cytokine is strongly associated with the most common symptom of primary Sjögren’s syndrome, the loss of salivary gland function. This finding led us to develop a novel ther- apy in the treatment of Sjögren’s syndrome by increasing the water permeability of the gland to restore saliva flow. Our study demon- strates that the targeted increase of gland permeability not only resulted in the restoration of secretory gland function but also re- solved the hallmark salivary gland inflammation and systemic inflam- mation associated with disease. Secretory function also increased in the lacrimal gland, suggesting this local therapy could treat the sys- temic symptoms associated with primary Sjögren’s syndrome.

Project description:Despite being one of the most common rheumatologic diseases, there is still no disease-modifying drug for primary Sjögren’s syndrome (pSS). Advancing our knowledge of the target tissue has been limited by the low dimensionality of histology techniques and the small size of human salivary gland biopsies. The goal of this study was to characterize the gene expression profile of total B cells, CD4+ T cells and epithelial cells in the target tissue of the most commonly-used model of pSS (the NOD model of pSS characterized by severe dacryoadenitis developing in males between 8 and 16 weeks of age).

High-throughput sequencing of IgH gene in minor salivary glands from Sjögren’s syndrome patients reveals dynamic ectopic lymphoid structures with recirculating B cells

Project description:This study is the first of this kind performed on minor salivary glands (mSG) of patients with Sjögren’s Syndrome. It shows that B cells clones are able to circulate across different glands and once re-seeded in a different mSG to continue the process of somatic hypermutation and switching of the immunoglobulin genes. Understanding B cells behavior in organs affected by autoantibodies, may help in designing personalized therapies, using monoclonal antibodies that target B cells (e.g. Rituximab) only on those patients that have a higher chance to respond.

Transcription profiling by array of parotid tissue from Primary Sjögren’s Syndrome and controls

Project description:Primary Sjögren’s syndrome (pSS) is a chronic autoimmune disease with complex etiopathogenesis. Here we use Affymetrix U133 plus 2.0 microarray gene expression data from human parotid tissue. Parotid gland tissues were harvested from 17 pSS and 14 14 non-pSS sicca patients and 18 controls. The data were used in the following article: Nazmul-Hossain ANM, Pollard RPE, Kroese FGM, Vissink A, Kallenberg CGM, Spijkervet FKL, Bootsma H, Michie SA, Gorr SU, Peck AB, Cai C, Zhou H, Horvath S, Wong DTW (2012) Systems Analysis of Primary Sjögren’s Syndrome Pathogenesis in Salivary Glands: Comparative Pathways and Molecular Events in Humans and a Mouse Model. Parotid gland tissues were harvested from 17 pSS and 14 non-pSS sicca patients and 18 controls.

Salivary gland transcriptomic analysis in the IL-14α transgenic mouse model of Sjögren’s disease using Visium V1 3' spatial gene expression assay

Project description:Sjögren’s disease (SjD) is a systemic autoimmune disorder primarily affecting the exocrine glands and characterized by dry mouth and dry eye, the presence of anti-SSA and/or anti-SSB autoantibodies in blood serum, and chronic lymphocytic infiltration of salivary and lacrimal glands (i.e., sialadenitis and dacryoadenitis, respectively). The interleukin-14α transgenic (IL-14αTG) mouse model of SjD recapitulates many aspects of human SjD, including progressive sialadenitis, loss of salivary gland function, and development of B cell lymphoma. We performed Visium V1 3' spatial transcriptomic analysis of fresh frozen submandibular and sublingual glands from a 12-month-old IL-14αTG Sjögren’s-like mouse and age-matched C57BL/6 control mouse.

Project description:In this study we have explored the effects of ERdj5 chaperone protein deletion in the proteome of 129SV mouse submandibular salivary glands. This knockout mouse model recapitulates many aspects of Sjögren’s Syndrome, including salivary gland inflammatory infiltrations, anti-nuclear autoantibodies and others (Apostolou et al., Front.Immunol.2019;10,506). Additionally, for 12SV wildtype animals we explore the proteome profile of the extensive sexual dimorphism observed in the submanidular glands of mice.

Identification of M3 muscarinic acetylcholine receptor reactive Th17 cells in patients with primary Sjögren’s syndrome

Project description:Previously M3 muscarinic acetylcholine receptore (M3R) reactive Th1 cells were identified, and here, M3R reactive Th17 cells were identified in peripheral blood of primary Sjögren’s syndrome patients using ELISpot method. To assess TCR repertoire overlap between identified M3R reactive Th17 cells, and salivary gland infiltrating T cells, we performed high throughput TCR sequencing of those cells from pSS patient.
