Hypercholesterolemia interferes with the cardioprotective and hidden cardiotoxic effects of rofecoxib

Project description:Rofecoxib, a cyclooxygenase-2 enzyme inhibitor, was withdrawn from the market after revealing its proarrhythmic and prothrombotic effects in phase IV clinical trials. Currently, in the preclinical phase of drug development, the cardiac side effects are tested on healthy tissues or small animals. Metabolic comorbidities, like hypercholesterolemia, may interfere with the cardioprotective or carditoxic effects of specific drugs. Here, we aimed to develop a test platform to investigate the cardiac effects of drugs in models with metabolic comorbidities. We fed adult, male Wistar rats for 12 weeks of high-fat diet (2% cholesterol + 0.25% cholic acid-enriched chow) or standard chow and treated high-fat diet-fed rats with 5.12 mg/kg rofecoxib for the last 4 weeks of the diet. Then, we performed RNA-seq from the left ventricles of the animals, to investigate the gene expression changes following high-fat diet and rofecoxib treatments.

Migraine-associated mutation in the Na,K-ATPase leads to disturbances in cardiac metabolism and reduced cardiac function

Project description:Background. Mutations in ATP1A2 gene encoding the Na,K-ATPase α2 isoform is associated with familial hemiplegic migraine type 2 (FHM2). Migraine with aura is a known risk factor for heart disease. The Na,K-ATPase is important for cardiac function but its role for heart disease remains unknown. We hypothesized that ATP1A2 is a susceptibility gene for heart disease and aimed to assess the underlying disease mechanism. Methods and Results. Mice heterozygous for the FHM2-associated G301R mutation in the Atp1a2 gene (α2+/G301R mice) and matching wild type (WT) controls were compared. Reduced expression of the Na,K-ATPase α2 isoform and increased expression of the α1 isoform was observed in hearts from α2+/G301R mice (Western blot). Left ventricular dilation and reduced ejection fraction was shown in hearts from 8-month-old α2+/G301R mice (cardiac magnetic resonance imaging) and this was associated with reduced nocturnal blood pressure (radiotelemetry). Cardiac function and blood pressure of 3-month-old α2+/G301R mice were similar to WT mice. Amplified Na,K-ATPase-dependent Src/Ras/Erk1/2 signaling was observed in hearts from 8-month-old α2+/G301R mice and this was associated with mitochondrial uncoupling (respirometry), increased oxidative stress (malonedialdehyde measurements), and a heart failure-associated metabolic shift (hyperpolarized magnetic resonance). Mitochondrial membrane potential was similar between the groups (JC-1 dye assay). Proteomics of heart tissue further suggested amplified Src/Ras/Erk1/2 signaling and increased oxidative stress and provided the molecular basis for systolic dysfunction in 8-month-old α2+/G301R mice. Conclusions. Our findings suggest that ATP1A2 mutation leads to disturbed cardiac metabolism and reduced cardiac function mediated via Na,K-ATPase-dependent ROS signaling through the Src/Ras/Erk1/2 pathway.

Hippocampal Transcriptomic Signatures of Stress Resilience and Susceptibility in the CSDS Mouse Model 7 hours after the last stress

Project description:This RNA-seq experiment investigates hippocampal transcriptional responses to chronic social stress in mice, aiming to uncover molecular signatures of behavioral resilience and susceptibility. Male C57BL/6J mice were subjected to a 10-day Chronic Social Defeat Stress (CSDS) protocol. At the end of the paradigm, animals were stratified into resilient (RES) and susceptible (SUS) groups using a dual behavioral index: the Social Ratio, which incorporates both interaction time and avoidance behavior in the presence of a novel CD1 aggressor. Hippocampal tissue was collected 7 hours after the final defeat session to capture transcriptional changes associated with the adaptive or maladaptive response to stress. RNA was extracted and processed for bulk RNA-seq on the Illumina platform. Analysis of differentially expressed genes revealed a marked transcriptional asymmetry in SUS mice, characterized by a widespread and selective upregulation of activity-dependent and stress-responsive genes. This dataset enables the exploration of gene expression dynamics and alternative splicing events underlying individual differences in stress adaptation.
