Single-Nucleus Multi-Omics Analysis of Adhesive Planar Retinal Organoids Derived from Self-formed Ectodermal Autonomous Multi-zone (SEAM)
Ontology highlight
ABSTRACT: We developed novel adhesive and planar retinal organoids from self-formed ectodermal autonomous multi-zone (SEAM), which recapitulates early human eye development. These organoids can be generated using a simple and reproducible method and are highly compatible with imaging techniques. The organoids exhibit stratified retinal architecture, contain all major retinal cell types, and feature mature photoreceptors. Retinal organoids at 12 and 36 weeks of differentiation were dissociated with Accutase, and nuclei and gene expression libraries were prepared using the Chromium Next GEM Single Cell Multiome ATAC + Gene Expression Reagent Kits, following the manufacturer’s protocol.
