Project description:RNA-seq of single mouse embryos using SMART-seq2. The embryos have been generated with IVF and are staged as hours post-IVF. At the zygote stage (~ 4h post-IVF), the embryos have been microinjected with 1.7 μg/μL Kdm5b wild-type (WT) or catalytic mutant (CM) mRNA. Non-injected embryos are denoted as “Noninj”. The embryos were cultured until collection at 20 h, 23 h, and 26 h post-IVF. A note on replicates: The replicate annotation refers to the number of times the specific condition was collected, i.e., the N number. It does not reflect the experimental batch. For the IVF batch, please refer to the sample sheet provided in the accompanying Git repository.

High Resolution Mapping of Mouse Embryonic Genome Activation using SMART-seq2 with in vitro polyadenylation

Project description:Single-embryo SMART-seq of mouse Embryonic Genome Activation with 2-cell stage embryos collected after 2 h precision in vitro fertilization (IVF) at the following timepoints: 17, 20, 23, and 26 h post-IVF. Individual embryos were lysed in total cell lysis buffer, and the lysates were split to perform in vitro polyadenylation on half of the sample. SMART-seq2 libraries were generated subsequently. All samples have matched in vitro polyadenylated (PolyA+) and nonadenylated (PolyA-) libraries.

SMART-seq2 of Kdm5b Overexpression in Fully Grown Oocytes

Project description:RNA-seq of single Fully Grown Oocytes using SMART-seq2. The oocytes were collected from female FVB mice, approximately. 44-48 h post-PMSG injection. The oocytes were microinjected with 1.3-1.7 μg/μL Kdm5b wild-type (WT) or catalytic mutant (CM) mRNA. Noninjected oocytes are denoted as “Noninj”. The oocytes were cultured overnight for approximately 16 h in M2 containing 20 μM Milrinone and 5% FBS and collected for sequencing.

Project description:This study includes raw data of blastocyst single-embryo SMART-seq profiles generated by IVF using sperm derived from mature (50-57 weeks) and young (9-11 weeks) males of C57BL/6N background. After SMART-seq library preparation of single embryos, indexed samples were pooled and sequenced on the NextSeq 2000 using a P3 flow cell in 50bp paired-end mode, targeting an average of 5 million reads per sample.
