RNA-Seq of FACS sorted Enteric Glia cells from the LMMP after T-cell activation via CD3

Project description:The Experiment was performed to understand the changes in expression profile of enteric glia cells in context of intestinal inflammation. Enteric glia are crucial regulators of gastrointestinal motility, however enteric glia alterations in IBD are poorly characterized. Acute T-cell activation rapidly impacts enteric glia activation, and induces cell death pathways, which negatively impacts small intestinal transit. This experiment has been crucial to identify IFNγ- and TNFα -driven necrosis in activated glia cell subsets. In order to sequence pure enteric glia, a mouse strain of Bl6 mice with a Plp1CreERT insert for glial specificity crossed with mice with a flox controlled tdTomato fluorescent protein on the ROSA26 Locus. The mice received a peritoneal injection of an anti-CD3 antibody or its isotype control antibody. At 6 hours, the small intestine was collected and the longitudinal muscle layer with the myenteric plexus stripped off. After enzymatic dissociation and trituration, the enteric glia were FACS sorted and finally the RNA isolated.

Project description:UV-treated cells were stained in green and seeded in density-matched monoculture with equally treated cells or in co-culture with untreated cells. The cultures were kept for 12 h prior to cell sorting and RNA isolation for bulk RNA sequencing. We wanted to compare transcriptional signatures of the UV-treated cells incubated in co-culture with the control set kept in monocultre, because the two cellular populations dispalyed morphologically different response to UV.

Identification of the late endosome LE/MVB proteome related to the endosomal microautophagy eMI activity in the rat liver.

Project description:To identify the late endosome LE/MVB proteome related to the endosomal microautophagy eMI activity we purified subcellular organelles including cytosol, late endosomes (LE/MVBs) and lysosomes (CMA+) from the rat liver and performed co-Ip experiment with antibodies directed against the major cellular chaperones mediating CMA and eMI. As such, we analyzed Hsc70-interacting proteins in LE/MVB, CMA+ lysosomes and cytosol by performing pull-down experiments (co-IP) using anti-Hsc70 antibodies. A comparative analysis of the Hsc-70 interactomes revealed that proteins bound to Hsc70 in cytosol and LE/MVBs or in cytosol and CMA+ lysosomes were considered eMI or CMA substrates, respectively.

Characterisation and use of orthotopically grownHapT1tumours in its homologous host as a preclinical model to evaluate novel drugs against desmoplastic pancreatic cancer

Project description:The major objective of this study was to characterise theHapT1 orthotopic hamster pancreatic tumor as a preclinical model of desmoplastic pancreatic ductal adenocarcinoma; and use this model to validate the efficacy of drugs that kills activated pancreatic stellate cells (PSCs)in vitro. Experimentaldesign: Commercially procured HapT1 pancreatic cancer (PCA) cell line was implanted in the pancreas of its syngeneic host, Syrian golden hamster (Mesocricetusauratus). After certain time period the primary and secondary tumors were harvested for histological andimmunophynotypical analysis. PSCs of hamsters were harvested, cultured and characterised. The in-cultured activated rodent PSCs and commercially procured human PSCs were used to check the cytotoxic effect of Disulfiram (DSF) in presence and absence of copper (Cu )in vitro. Finally, theHapT1 orthotopic tumor model was used to check the efficacy of DSF in vivo.
