Mesaconate Attenuates LPS/D-GalN-Induced Acute Liver Failure by Enhancing HIF-1α-Mediated AXL-Dependent Efferocytosis

ABSTRACT: Mouse bone marrow-derived macrophages were extracted from the tibia, fibula and femur of C57BL/6J mice and cultured in DMEM medium containing 10% fetal bovine serum (FBS) (Gibco), 1% penicillin-streptomycin and 20ng/ml M-CSF for 7 days. BMDM was either continuously treated with 10ng/ml LPS for 7 hours or pretreated with 10 mM ITA or Mesa for 4 hours, followed by stimulation with 10 ng/mL LPS for 3 hours. After that, the culture medium was discarded and 1ml of RNAiso Plus was added to each well. The samples were stored at -80℃ and quantitatively analyzed using the sequencing platform of the Beijing Institute of Genomics.
