Effect of hepatocyte-restricted deletion of fgf21 deletion on liver gene expression in response to fasting in mice

ABSTRACT: FGF21 is a liver-derived hormone potently induced in response to fasting. However, its role in the regulation of hepatic gene expression is unknown. The effect of 20h fasting on liver gene expression was investigated in wild-type mice and in mice lacking hepatocyte FGF21. Fgf21 hepatocyte-specific knockout (Fgf21hep-/-) mice were generated by mating the floxed-Fgf21 mouse strain (B6.129S6(SJL)Fgf21 < tm1.2Djm>/J provided by The Jackson Laboratory) with C57BL/6J albumin-Cre transgenic mice, to obtain albumin-Cre+/-Fgf21flox/flox mice, as described previously (Fougerat et al., 2022). Albumin-Cre-/-Fgf21flox/flox (Fgf21hep+/+) littermates were used as controls. 13-week-old male mice were fed ad libitum or fasted for 20h (starting at ZT0) (n = 5-6 male mice/genotype/experimental condition).
