Effect of hepatocyte FGF21 on brown adipose tissue gene expression in mice

ABSTRACT: FGF21 is an hepatokine, a liver-derived hormone, that is highly induced and secreted in response to fasting. We investigated the role of hepatic FGF21 on gene expression profile in the brown adipose tissue. The effect of 20h fasting on brown adipose tissue gene expression was investigated in male wild-type mice and in mice lacking hepatocyte FGF21. Fgf21 hepatocyte-specific knockout (Fgf21hep-/-) mice were generated by mating the floxed-Fgf21 mouse strain (B6.129S6(SJL)Fgf21 < tm1.2Djm>/J provided by The Jackson Laboratory) with C57BL/6J albumin-Cre transgenic mice, to obtain albumin-Cre+/-Fgf21flox/flox mice, as described previously (Fougerat et al., 2022). Albumin-Cre-/-Fgf21flox/flox (Fgf21hep+/+) littermates were used as controls. 13-week-old male mice were fed ad libitum or fasted for
