RNA-seq data from a differentiation time course of primary human preadipocytes

ABSTRACT: RNA-seq files collected from an in vitro differentiation time course of primary human adipocytes from lean, obese, and obese with T2D individuals. RNA-seq data was collected at confluence (day 0), day 3, and day 15 of differentiation. \"Conf\" samples are preadipocytes, \"3days\" and \"15days\" are differentiated adipocytes.

Tear proteins altered in patients with persistent eye pain after refractive surgery: biomarker candidate discovery

Project description:Some patients develop persistent eye pain after refractive surgery, but the factors that cause or sustain pain are unknown. We tested whether tear proteins of patients with pain 3 months after surgery differ from patients without pain. Patients undergoing refractive surgery (LASIK or PRK) were recruited from 2 clinics and tears were collected 3 months after surgery. Participants rated their eye pain using a numerical rating scale (NRS, 0 – 10; no pain – worst pain) at baseline, 1 day, and 3 months after surgery. Using tandem mass tag (TMT) proteomic analysis, we examined tears from participants with NRS > 3 at 3 months (n=16) and a comparator group with NRS < 1 at 3 months after surgery (n=32). A subset of proteins (83 of 2748 detected, 3.0%) were associated with pain 3 months after surgery. High-dimensional statistical models showed that magnitude of differential expression was not the only important factor in classifying tear samples from pain patients. Models utilizing 3 or 4 proteins had better classification performance than single proteins and represented differences in both directions (higher or lower in pain). Thus, patterns of protein differences may serve as biomarkers of post-surgical eye pain as well as potential therapeutic targets.

SH2B1 Promotes Apoptosis in Diabetic Cataract via p38 MAPK Pathway

Project description:Diabetic patients face an increased risk of developing cataracts, with unclear mechanisms. Our study illuminates these mechanisms by identifying differentially expressed proteins in the lens anterior capsule of diabetic cataract (DC) and age-related cataract patients using quantitative proteomics. We found SH2B1 to be crucial in DC progression. Reduced SH2B1 expression was confirmed through PCR and Western blotting in patient samples, diet-induced obese mice, and high-glucose (HG)-cultured human lens epithelial cells. Under HG conditions, cell proliferation decreased, while migration and apoptosis, alongside changes in Bcl2 and caspase 3 expression, increased. Overexpressing SH2B1 alleviated these changes and influenced the p38 MAPK signaling pathway. This suggests SH2B1 and the p38 MAPK pathway as significant in DC pathogenesis and potential therapeutic targets. Clinically, this could lead to therapies aimed at halting or slowing DC progression.

Project description:Bone marrow lesions (BML) are well described in osteoarthritis (OA) using magnetic resonance imaging (MRI) and associate with pain however, little is known about their role in disease process and pattern of gene expression within the lesions. This study evaluated the gene expression profile of OA BML (n=14) in comparison to normal bone (n=10) by microarray profiling. MR imaging and the MOAKS scoring was used to locate lesions and the scaled axial images were used to sample BMLs. The bone tissue controls were harvested from participants undergoing surgery following trauma.

Human Ovarian Proteome - A draft map of the human ovarian proteome for tissue engineering and clinical applications

Project description:Fertility preservation research in women today is increasingly taking advantage of bioengineering techniques to develop new biomimetic materials and solutions to safeguard ovarian cell function and microenvironment in vitro and in vivo. However, available data on the human ovary are limited and fundamental differences between animal models and humans are hampering researchers in their quest for more extensive knowledge of human ovarian physiology and key reproductive proteins that need to be preserved. We therefore turned to multi-dimensional label-free mass spectrometry to analyze human ovarian cortex, as it is a high-throughput and conclusive technique providing information on the proteomic composition of complex tissues like the ovary. In-depth proteomic profiling through two-dimensional liquid chromatography-mass spectrometry, western blot, histological and immunohistochemical analyses, and data mining helped us to confidently identify 1,508 proteins. Moreover, our method allowed us to chart the most complete representation so far of the ovarian matrisome, defined as the ensemble of extracellular matrix proteins and associated factors, including more than 80 proteins. In conclusion, this study will provide a better understanding of ovarian proteomics, with a detailed characterization of the ovarian follicle microenvironment, in order to enable bioengineers to create biomimetic scaffolds for transplantation and three-dimensional in vitro culture. By publishing our proteomic data, we also hope to contribute to accelerating biomedical research into ovarian health and disease in general
