Tie2+ vs Tie2- macrophages bulk RNAseq

ABSTRACT: BulkRNAseq to identify Tie2+ macrophages signature. Cells were sorted for CD14+ and Tie2 (PTEK). Two fractions were hence sequenced on NextSeq platform (Illumina) after full length cDNA amplification using the SMART-Seq mRNA LP kit (Takara).

miRNA expression profiles of TIE2 expressing Monocytes (TEMs)

Project description:To identify the miRNA expressing profiles of TIE2 expressing Monocytes (TEMs), we have employed the Agilent Human miRNA 8×60K (Design ID:046064) microarray. Human peripheral blood mononuclear cells (PBMCs) in venous blood from healthy donors were isolated by Lymphoprep (Axis-Shield, Norway). Human monocytes in PBMCs, identified as cells that expressed CD14, were enriched by positive immunomagnetic selection using anti-CD14 MicroBeads (Miltenyi, Germany). TEMs (TIE2+CD14+) and TIE2-Monocytes (Tie2-CD14+) were then isolated by FACS-sorting (Aria II, BD Biosciences) using FITC-conjugated anti-CD14 (BD Biosciences, USA) and APC-conjugated anti-TIE2 (R&D System, USA) antibodies.Three TEMs samples together with their paried TIE2-Monocytes were detected.

The gene expressions of three independent paried TEMs and TIE2- Monocytes samples from different donors were measured.

Project description:Comparison of transcriptional profiles of human umbilical vein endothelial cells (HUVECs) expressing wild-type vs. VM-causative mutant forms of TIE2/TEK. The effects of the most common Venous Malformation-causative mutations in the endothelial cell tyrosine kinase receptor: R849W and L914F, were tested. 743 genes were differentially expressed across the four groups. The 80 genes distinguishing between L914F and wild-type TIE2-expressing HUVECs were analyzed in greater detail. 3 batches each of: non-transfected, and wild-type TIE2, R849W-TIE2, and L914F-TIE2 overexpressing HUVECs were compared by exon-array profiling.

Comparison of fibroblasts from ARH3-mutated patients and controls by transcription profiling

Project description:Mutations in the ARH3 (ADPRHL2) hydrolase that catabolises ADP-ribose are associated with neurodegeneration and ataxia. In our experiments we have found that ARH3-defective cells accumulate mono-(ADP-ribose) scars on histones that may prevent other epigenetic modifications from happening at these sites. Transcriptomic analyses thus could be useful to show whether and how persistence of these marks might translate into the pathogenesis. We have employed primary human fibroblasts (cultured in individual cell lines) from two ARH3-mutated patients (A1, A2), harbouring the homozygous mutations Q334* (A1 & A2/siblings) and two unaffected controls (ARH3 ctrl/father & unrelated 1BR) to run RNA-seq. Due to the sensitive nature of the data, raw data files have not been uploaded.

Tie2 regulates endocardial sprouting and myocardial trabeculation

Project description:The ang1-Tie2 pathway is required for normal vascular development, but its molecular effectors are not well-defined during cardiac ontogeny. Here we show that endocardial specific attenuation of Tie2 results in mid-gestation lethality due to heart defects associated with a hyperplastic but simplified trabecular meshwork (fewer but thicker trabeculae). Reduced proliferation and production of endocardial cells (ECs) following endocardial loss of Tie2 results in decreased endocardial sprouting required for trabecular assembly and extension. The hyperplastic trabeculae result from enhanced proliferation of trabecular cardiomyocyte (CMs), which is associated with upregulation of Bmp10, increased retinoic acid (RA) signaling, and Erk1/2 hyperphosphorylation in the myocardium. Intriguingly, myocardial phenotypes in Tie2-cko hearts could be partially rescued by inhibiting in utero RA signaling with pan-retinoic acid receptor antagonist BMS493. These findings reveal two complimentary functions of endocardial Tie2 during ventricular chamber formation: ensuring normal trabeculation by supporting EC proliferation and sprouting, and preventing hypertrabeculation via suppression of RA signaling in trabecular CMs.
