Xenografts and lung metastases derived from neuroblastoma cell lines overexpressing Anaplastic lymphoma kinase (ALK) variants engrafted in immunodeficient mice
Ontology highlight
ABSTRACT: Neuroblastoma cell lines genetically modified to overexpress Anaplastic lymphoma kinase (ALK) point mutations or the ALK wild-type isoform were injected subcutaneously or orthotopically into NOD-SCID-gamma or athymic Swiss nude mice, respectively. After development of the tumor, RNA was extracted from the tumor or lungs (metastases) and analyzed by bulk RNA-sequencing. The human neuroblastoma cell lines used are: SK-N-Be2C, SH-EP, or GIMEN. ALK genotype of the cell lines: wild type for SK-N-Be2C and GIMEN, mutation F1174L heterozygous for SH-EP. The vector used for transfection are: ALK-F1174L, ALK-R1275Q, ALK-wt (wild-type) or pLIV (empty vector). ALK-F1174L and ALK-R1275Q are the most common ALK point mutations observed in neuroblastoma tumors.
