MicroRNA profiling in human colon cancer cell lines

ABSTRACT: MicroRNA profiling in colon cancer cell lines. Two cell lines have been found with impairment on the export of precursor miRNAs into the cytoplasm (XPO5), HCT15 and DLD1. To test the microRNAs involved several comparisons have been performed: DLD1 and HCT15 cell lines have been compared with XPO5 wild-type cell lines (HCT116 and RKO). Furthermore the transfected XPO5 wild-type defective cells (DLD1.XPO5 wt and HCT15.XPO5 wt) have been compared respect to the non-transfected cells (HCT15 and DLD1). Finally the cell lines HCT15 and DLD1 transfected with a negative control (HCT15.XPO5 358-366 and DLD1.XPO5 358-366) have been compared with respect to non-transfected cell lines (HCT15 and DLD1).

MicroRNA profiling in four human bladder cancer cell lines

Project description:To identify miRNA-target interactions that are important in bladder cancer metastasis, we profiled miRNA and mRNA expression in poorly metastatic cell lines T24 and LUC, and their metastatic derivatives FL4 and LUL2, respectively. We reasoned that miRNAs and genes that expressed differently between parental and derivative cell lines could potentially contribute to their disparity in metastatic competence. We further hypothesized that interactions between these miRNAs and target genes may play an important role in metastatic competence. We have developed the multiMiR R pacakge and database for such integrated analysis of miRNA-target interactions. Here we demonstrate how multiMiR was used to generate testable hypotheses that could be pursued experimentally. The data set here is from the miRNA expression profiling experiment. microRNA profiles of EZH2 activation

Project description:Enhancer of Zeste Homolog 2 (EZH2) is a histone methyltransferase that catalyzes the trimethylation of histone H3 lysine 27 (H3K27me3). EZH2 expression is significantly upregulated in MPNST. In our study, we investigated the function of EZH2 and the molecular mechanisms that are regulated by EZH2 in MPNST pathogenesis. Our findings enhance the knowledge of EZH2’s function and biology, and have the potential to provide a novel therapeutic approach for MPNST patients in the clinic. An EZH2 knockdown experiment was carried out in MPNST cells. Radiotherapy-induced miR expression influences breast cancer recurrence formation.

Project description:The act of surgery strongly stimulated breast cancer cell growth in mice. Local irradiation of wounded breast efficiently prevented wounded-induced breast cancer cell growth. These modifications were linked to miR modulation both in human specimens and in the mouse model, in vivo. The most significantly IORT-induced miR was miR-223. miR-223 expression controlled EGF production and, eventually, cancer cell proliferation and survival.

Project description:To validate the suitability of two commonly used colorectal cancer cell lines, DLD1 and SW480, as model systems to study colorectal carcinogenesis, we treated these cell lines with β-catenin siRNA and identified β-catenin target genes using DNA microarrays. The list of identified target genes was compared to previously published β-catenin target genes found in the PubMed and the GEO databases. Based on the large number of β-catenin target genes found to be similarly regulated in DLD1, SW480 and LS174T as well as the large overlap with confirmed β-catenin target genes, we conclude that DLD1 and SW480 colon carcinoma cell lines are suitable model systems to study β-catenin regulated genes and signaling pathways
