Project description:Thylakoid membranes are the specialized internal membrane system produced in plants, algae, and cyanobacteria to convert sunlight to chemical energy via oxygenic photosynthesis. Cyanobacterial thylakoid membranes harbor the protein complexes and electron transport molecules that are necessary for photosynthetic light reactions and respiratory electron flow. The thylakoid membranes sit between the plasma membrane and the central cytoplasm, leading to intricate cellular compartmentalization. How thylakoid membranes are generated to form the functional network and how protein complexes are recruited into thylakoids remain elusive. Here, we developed a method to modulate thylakoid biogenesis in the model cyanobacterium Synechococcus elongatus PCC7942 and probed the spatial-temporal stepwise biogenesis process of cyanobacterial thylakoid membranes, using electron microscopy, in situ cryo-electron tomography, confocal microscopy, mass spectroscopy, and biochemical approaches. Our results revealed that the plasma membrane and regularly-arranged concentric thylakoid layers have no physical connections. The newly synthesized thylakoid membrane fragments commerce between the plasma membrane and pre-existing thylakoids, where the initial biogenesis of Photosystem II occurs. Photosystem I monomers appear in thylakoid membranes earlier than other photosystem assemblies. Redistribution of photosynthetic protein complexes during thylakoid biogenesis ensures establishment of the spatial organization of the functional thylakoid membrane network. This study provides insights into the molecular processes of the photosynthetic machinery biosynthesis and organization.

Project description:Light is the driving force of photosynthesis. However, excessive light can be harmful. Photoinhibition, or lightinduced photodamage, is one of the key processes limiting photosynthesis. When the absorbed light exceeds the amount that can be dissipated by photosynthetic electron flow and other processes, damaging radicals are formed that mostly inactivate photosystem II (PSII). A well defined mechanism that protects the photosynthetic apparatus from photoinhibition has been described in the model green alga Chlamydomonas reinhardtii and plants. Chlorella ohadii is a green microalga, isolated from biological desert soil crusts, that thrives under extreme high light (HL) in which other organisms do not survive. Here, we show that this alga evolved unique protection mechanisms distinct from those of C. reinhardtii and plants. When grown under extreme HL, significant structural changes were noted in the C. ohadii thylakoids, including a drastic reduction in the antennae and the formation of stripped core PSII, lacking its outer and inner antennae. This is accompanied by a massive accumulation of protective carotenoids and proteins that scavenge harmful radicals. At the same time, several elements central to photoinhibition protection in C. reinhardtii, such as psbS, the stress related light harvesting complex (LHCSR), PSII protein phosphorylation and state transitions are entirely absent or were barely detected in C. ohadii.

They were grown in a growth chamber for 5 weeks in short day (8/16 light/dark cycle) at 22 degrees, 150 umol*m2*s-1 (fluorescent lights) and 55% RH in 100-ml pots with Levingtons L2+S. On the day of the experiment, two hours after the lights turned on they were subjected to mild light stress (approx. 1500 umol*m-2*s-1, measured with a PAR light meter) for 30 min under an Isolight machine (white LED array), and immediately snap-frozen in liquid nitrogen. The outer leaves from four biological replicates were ground and RNA purified, and labelled RNA was produced with the Agilent Quick Amp kit. Labelled RNA was hybridized to Agilent Arabidopsis V4 microarray slides, and the fluorescence recorded with a Genepix scanner.

Project description:Poplar (Populus trichocarpa, clone Nisqually-1) plants were grown in a Conviron PGR 15 growth chamber using precise control of temperature, light, and humidity. Diurnal (driven) conditions included 12L:12D light cycles and 25C/12C thermocycles in three different combinations. These were: photocycles (LDHH), 12 hrs. light (L)/12 hrs. dark (D) at a constant temperature (25C; HH); photo/thermocycles (LDHC): 12 hrs. light (L) /12 hrs. dark (D) with a high day temperature (25C) and a low night temperature (12C); and thermocycles (LLHC): continuous light (LL) with 12 hrs. high/12 hrs. low temperature (25C, day; 12C, night). Light intensity and relative humidity were 700 micromol m-2s-2 and 50%, respectively. Three-month-old poplar plants were entrained for at least one week under the respective condition prior to initiation of each experiment. Leaves and stems from individual poplar plants were collected every four hours for 48 hrs in driven (diurnal) conditions followed by a two day freerun spacer under continuous light/temperature followed by two additional days of sampling under the same continuous free run condition.
