Microarray analysis of dengue seronegative or seropositive volunteers that received the tetravalent dengue vaccine TAK-003

Project description:Samples were collected from the DEN210 study, a phase IIb clinical trial evaluating two doses of TAK003, a tetravalent dengue vaccine, administered 90 days apart in healthy adult participants with and without prior DENV infection (NCT03746015). RNA was extracted from whole blood collected in Paxgene tubes from volunteers at Day 1, 4, 6, 90, 94 and 96. Day 1 and Day 90 samples were collected prior to administration of first and second TAK-003 vaccine dose. We performed gene expression analysis in dengue seronegative and seropositive individuals over time.

Differential miRNome analysis in a dengue resistant cohort versus susceptible after Dengue Virus serotype 2 or mock in vitro infection

Project description:MicroRNAs have emerged as relevant players in resistance to infection. We aimed to identify miRNAs involved in natural resistance to dengue infection. Two groups of people living in dengue endemic area were classified as susceptible (SD) or resistant (RD) to dengue virus (DENV) infection according to their anti-DENV antibody status. We hypothesized that anti-DENV seronegative individuals may represent a population resistant to DENV infection. The miRNome from monocytes of 7 individuals of each group was assessed upon mock- or DENV-2 infection.Dysregulated miRNAs were chosen as candidates for functional studies.

The RNA from 0h-post LPS stimulation were used as reference sample. Were also performed dye-swaps of each biological sample as technical replicates.

Live Attenuated Tetravalent Dengue Vaccination in Non-Human Primates

Project description:We measured genome-wide transcript abundance patterns in 246 whole blood samples collected from 35 cynomolgus macaques before and after vaccination with a live attenuated tetravalent dengue vaccine (TDV) or PBS (placebo). Animal work was conducted at the Charmany Instructional Facility of the School of Veterinary Medicine, University of Wisconsin-Madison, and all animal procedures were approved by the University of Wisconsin-Madison's Animal Care and Use Committee. Thirty-five adult male, DENV-seronegative cynomolgus macaques from Vietnam were placed in quarantine for 30 days prior to the start of the study. Five groups of animals (n=6 animals per group) received TDV either subcutaneously (SC) in 0.5 mL inocula or intradermally (ID) in 0.1 mL inocula using a needle-free injector (PharmaJet' device) or needle and syringe (N&S). Animals received two doses (same route) of TDV either on the same day (Day 0) or sixty days apart. One group (n=5) received PBS by ID route and needle-free injector. On day 90 after primary vaccination, 3 animals from each group were challenged with 105 PFU wt DENV-2 (New Guinea C strain) or 105 PFU wt DENV-4 (Dominica/81 strain) by SC route using N&S. Whole blood samples were collected for transcriptional profiling from all animals on days -11, -2, 1, 3, 5, and 7 before and after immunization, and for placebo animals on days 88, 91, 93, 95, and 97 before and after wt DENV challenge. Blood samples were drawn into PAXgene Blood RNA tubes and stored at -80'C prior to RNA extraction and processing.

Transcription profiling of human cell types infected with Dengue virus reveals TRAIL Is a Novel Antiviral Protein against Dengue Virus

Project description:Dengue fever is an important tropical illness for which there is currently no virus-specific treatment. To shed light on mechanisms involved in the cellular response to dengue virus (DV), we assessed gene expression changes, using Affymetrix GeneChips (HG-U133A), of infected primary human cells and identified changes common to all cells. The common response genes included a set of 23 genes significantly induced upon DV infection of human umbilical vein endothelial cells (HUVECs), dendritic cells (DCs), monocytes, and B cells (analysis of variance, P < 0.05). Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), one of the common response genes, was identified as a key link between type I and type II interferon response genes. We found that DV induces TRAIL expression in immune cells and HUVECs at the mRNA and protein levels. The induction of TRAIL expression by DV was found to be dependent on an intact type I interferon signaling pathway. A significant increase in DV RNA accumulation was observed in anti-TRAIL antibody-treated monocytes, B cells, and HUVECs, and, conversely, a decrease in DV RNA was seen in recombinant TRAIL-treated monocytes. Furthermore, recombinant TRAIL inhibited DV titers in DV-infected DCs by an apoptosis-independent mechanism. These data suggest that TRAIL plays an important role in the antiviral response to DV infection and is a candidate for antiviral interventions against DV. We used Affymetrix microarrays to study the response of human host cells to dengue virus (DV). Experiment Overall Design: For three human cell types, RNA was extracted and hybridized on Affymetrix microarrays. We compared a total of 10 samples. Five were infected in vitro for 48 hours with DV, including HUVECs (n=2), monocytes (n=2), and B-cells (n=1). Five were mock-infected controls of the same cell types and numbers. From these samples, were identified 23 genes that were induced by DV infection in all of the cell types.
