Bulk RNA-seq of murine G-MDSCs after co-culture with ILC2s and control G-MDSCs

ABSTRACT: This experiment aimed to characterize the transcriptional response of murine granulocytic myeloid-derived suppressor cells (G-MDSCs) upon interaction with group 2 innate lymphoid cells (ILC2s). G-MDSCs were isolated from the bone marrow of C57BL/6 mice, while ILC2s were induced in vivo by hydrodynamic gene transfer of IL-25, IL-33, and IL-9 expression plasmids, followed by isolation from spleen and mesenteric lymph nodes. Purified G-MDSCs were co-cultured with ILC2s using a transwell system that allowed exchange of soluble factors without direct cell contact. After 48 hours, G-MDSCs were collected for bulk RNA sequencing and compared to G-MDSCs cultured alone to identify ILC2-induced transcriptional changes. The dataset provides insight into cytokine-mediated crosstalk between G-MDSCs and ILC2s and its potential role in immune regulation and tissue remodeling.
