RNA seq of TCR75 cells following murine heart transplantation, tolerance induction, and immune challenge

ABSTRACT: CD4+CD45.1+ TCR75/Rag-/- T cells (TCR75) were adoptively transferred into CD45.2+ B6 or Foxp3-DTR hosts prior to transplantation of a B/c heart (HTx) in the presence or absence (Rej) of anti-CD154 (day 0, day 7, day 14) and DST (day 0) to induce tolerance. A group of tolerant mice were depleted of Tregs with diphtheria toxin on d35 post-transplantation (Treg Depl.) or injected with plasmids encoding IL-6 and IFNb (Plasmid Inj.). CD45.1+ TCR75 cells were harvested from the spleen and lymph nodes on d40 post-transplantation for Treg Depl. and Plasmid Inj, or d5 post-transplantation in Rej.,enumerated and subjected to bulk RNAseq.
