RNA-seq of mouse intestinal adenoma organoids treated with pre- and post-exercise human serum

ABSTRACT: This experiment investigated transcriptomic responses of mouse intestinal adenoma organoids to circulating factors induced by acute exercise. Intestinal adenoma organoids derived from an Lgr5-creER;Apcfl/fl mouse model were cultured with 1% human serum collected immediately before or after a maximal incremental cycling exercise test. Paired pre- and post-exercise serum samples from six participants, comprising three younger and three older adults, were used to stimulate organoids for five days. Total RNA was then isolated, poly(A)-enriched mRNA libraries were prepared and sequenced on an Illumina platform. Reads were aligned to the Mus musculus GRCm38 reference genome and transcript abundance was quantified. Differential expression and gene-set enrichment analyses were used to examine tran
