CD27 signaling in CD8+ T cells in vivo

ABSTRACT: Wild type or Cd27-/- mice of 8-12 weeks old were immunized intranasally with 500 ug ovalbumin (OVA) protein and 1 ug cholera toxin in 50 ul HBSS. Spleen, draining lymph node (DLN) or lung were isolated at day 3, day 4 or day 8 after immunization. Single cell suspensions of indicated organs were prepared and OT-I T cells were isolated from this using flow cytometric cell sorting.

Various cell types can produce the chemokine CXCL10 in response to IFN-γ stimulation. CXCL10 is generally viewed as a proinflammatory chemokine that promotes recruitment of CD8÷ and Th1-type CD4÷ effector T cells to infected or inflamed nonlymphoid tissues. We show that CXCL10 plays a role during CD8÷ T cell priming in the mouse. Project description:Using gene expression profiling of mammary tumors generated in a mouse model for BRCA1-associated breast cancer we have searched for markers that correlate with response to the anti-cancer drugs docetaxel or cisplatin. To validate the results, the analysis was done on 2 different platforms (39K Mouse Exonic Evidence Based Oligonucleotide (MEEBO), and 45K Illumina Sentrix mouse V6 single-channel bead arrays). Moreover, we analyzed mouse mammary tumors that had acquired docetaxel resistance on the MEEBO platform.

Project description:The objective of the current study was to evaluate the performance of frequently used E. coli K-12 strains HMS174 and RV308 in the context of industrial cultivations. To meet this objective, we carried out batch cultivations with high-glucose media in computer-controlled bioreactors and applied transcriptome analysis to interpret phenotypic attributes at the molecular level. The resulting enhancement of knowledge about host physiology is expected to contribute to the acceleration of improvements and optimizations in related biotech processes.

Transcription profiling of E. coli HMS174(DE3)(pET30aNproGFPmut3.1) to analyze cellular response to limited induction with IPTG

Project description:This transcription profiling time course experiment was conducted in order to analyze the cellular response of the T7 RNA polymerase based E. coli expression system HMS174(DE3)(pET30aNproGFP) to a limited induction with isopropyl-beta-D-galactoside (IPTG). The used cell material was produced under well controlled and defined conditions in an exponential carbon limited fed-batch cultivation at a growth rate of 0.1 per hour. One doubling past feed start induction was started by continuously feeding limiting amounts of inducer gaining in a constant IPTG to cell dry weight ratio of 1µmol/g. Due to the low induction level a physiologically tolerable recombinant gene expression rate was generated and production period was significantly elongated. Non induced cells sampled at the time point of induction were used as reference and compared to cells subsequently sampled during the production period (sampling frequency 2 hours).

Automated array-CGH optimized for archival formalin-fixed, paraffin-embedded tumor material

Project description:BACKGROUND: Array Comparative Genomic Hybridization (aCGH) is a rapidly evolving technology that still lacks complete standardization. Yet, it is of great importance to obtain robust and reproducible data to enable meaningful multiple hybridization comparisons. Special difficulties arise when aCGH is performed on archival formalin-fixed, paraffin-embedded (FFPE) tissue due to its variable DNA quality. Recently, we have developed an effective DNA quality test that predicts suitability of archival samples for BAC aCGH. METHODS: In this report, we first used DNA from a cancer cell-line (SKBR3) to optimize the aCGH protocol for automated hybridization, and subsequently optimized and validated the procedure for FFPE breast cancer samples. We aimed for highest throughput, accuracy, and reproducibility applicable to FFPE samples, which can also be important in future diagnostic use. RESULTS: Our protocol of automated array-CGH on archival FFPE ULS-labeled DNA showed very similar results compared with published data and our previous manual hybridization method. CONCLUSION: This report combines automated aCGH on unamplified archival FFPE DNA using non-enzymatic ULS labeling, and describes an optimized protocol for this combination resulting in improved quality and reproducibility. In this study, we optimized the BAC araay-CGH protocol for automated hybridization for FFPE breast cancer samples. We have tested hybridization temperature and duration, different hybridization buffer conditions, and post-hybridization washing.
