Transcription profiling of Acanthamoeba polyphaga mimivirus infecting Acanthamoeba castellanii for different lengths of time

ABSTRACT: A culture of Acanthamoeba castellanii cells was infected with mimivirus. At specified times after infection, infected cells aliquots were subjected to total RNA extraction. Cy3 labeled cDNA was hybridized on custom designed Agilent oligonucleotide tiling microarrays covering both strands of the mimivirus genome.

Mimivirus, a virus infecting Acanthamoeba, is the prototype of the Mimiviridae, the latest addition to the nucleocytoplasmic large DNA viruses. The Mimivirus genome encodes close to 1000 proteins, many of them never before encountered in a virus, such as four amino-acyl tRNA synthetases.
