Polymicrobial periodontal pathogens transcriptomes in calvarial bone and soft tissue

Project description:The objectives of this investigation were to examine changes in the host transcriptional profiles during a polymicrobial periodontal pathogens Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia infection using a murine calvarial model of inflammation and bone resorption. P. gingivalis FDC 381, T. denticola ATCC 35404, and T. forsythia ATCC 43037 was injected into the subcutaneous soft tissue over the calvaria of BALB/c mice for 3 days, after which the soft tissues and calvarial bones were excised. RNA was isolated from infected soft tissues and calvarial bones and analyzed for transcript profiles using Murine GeneChipM-BM-. MG-MOE430A Affymetrix arrays to provide a molecular profile of the events that occur following infection of these tissues. We used mouse microarrays to detail the molecular profile of the events that occur following infection of calvarial and bone tissues and identified distinct classes of up- and down-regulated genes during this process. P. gingivalis/T. denticola/T. forsythia was injected at 5 x 108 each (N = 10) into the soft tissues overlying the calvaria of the mice for 3 days. A control group (N = 9) was injected with vehicle once daily for 3 days. Mice were euthanized 8 h after the last injection. The calvarial bones and overlying soft tissues from 5 mice in each group were excised, snap frozen in liquid nitrogen, and stored at M-bM-^@M-^S80M-BM-0C until RNA isolation. Total RNA was isolated from the frozen calvarial tissue and calvarial bone from each mouse (Polymicrobial bacteria infected and control animals, N = 5 in each group) with Trizol reagent (Invitrogen, CA). Equal amounts of RNA from samples were labeled and hybridized on a mouse GeneChip following the protocol described in the GeneChip Expression Analysis Technical manual (Affymetrix, Santa Clara, CA). After hybridization, the GeneChip arrays were stained and scanned in an Affymetrix GCS 3000 7G Scanner.

Project description:We utilized T. denticola as a model red complex organism to help determine whether red complex organisms can respond to one another upon cell-cell contact in mixed culture and the types of genes involved in such responses. We extracted RNA from combinations of T. denticola with either other P. intermedia, S. sanguinis, S. gordonii, F. nucleatum, P. gingivalis, or T. forsythia that had been centrifuged to induce cell contact, and identified the numbers and types of genes that were differentially regulated using microarrays.

Transcription profiling of Porphyromonas gingivalis wild type and mutant PG1432::ermF to identify genes which have their expression influenced by PG1432

Project description:The aim of this experiment is to ascertain which genes have their expression influenced by PG1432. PG1432 encodes a putative sensor histidine kinase which is part of two component signal transduction that was highly up-regulated in P. gingivalis mature biofilm cells. PG1432 was disrupted in P. gingivalis W50 by insertion of ermF cassette. The strain W50 PG1432 mutant was designated ECR222. Two independent replicates of each strain were grown for 30 days in continuous culture in chemostats using brain heart infusion media supplemented with 0.5 mg mL-1 cysteine and 5 ï¿½~Aï¿½g mL-1 hemin. Although strain W50 formed sufficient biofilm to use for total RNA harvest and microarray analysis, strain ERC222 formed only sparse biofilm showing that disruption of PG1432 impaired biofilm formation by this mutant. We were unable to collect sufficient biofilm cells of strain ECR222 therefore the microarray analysis total RNAs were harvested only from planktonic phase cells. The transcriptome of the mutant ECR222 were compared to that of strain W50.

Transcriptome response of Salmonella enterica subsp. enterica serovar Typhimurium N-15 and Bifidobacterium thermophilum RBL67 in co-culture

Project description:Bifidobacterium thermophilum RBL67 (RBL67), a human fecal isolate and promising probiotic candidate, showed antagonistic and protective effects against Salmonella and Listeria in vitro. However, the underlying mechanisms fostering these health-related effects remain unknown. Therefor the transcriptome response of RBL67 and Salmonella enterica subsp. enterica serovar Typhimurium N-15 (N-15) in co-culture compared to the response in their respective mono-cultures. RNA was extracted from culture samples taken after 4 (N-15) or 5 h (RBL67) and RNAseq was performed on an Illumina HiSeq 2000 sequencer. Three biological replciates were performed resulting in 12 data sets: 3 RBL67 mono culture, 3 N15 mono-culture, 3 RBL67 co-culture, 3 N15 co-culture. Our study provided first insights into probiotic-pathogen interaction on transcriptional level and suggests a mechanism for how probiotic organisms can protect the host from infections. RNA was extracted from culture samples taken after 4 (N-15) or 5 h (RBL67) and RNAseq was performed on an Illumina HiSeq 2000 sequencer. Three biological replciates were performed resulting in 12 data sets: 3 RBL67 mono culture, 3 N15 mono-culture, 3 RBL67 co-culture, 3 N15 co-culture.
