Project description:Obesity is well recognized as a risk factor for coronary heart disease and mortality. The relationship between abdominal obesity and ischemic stroke remains less clear. Previous publication showed the obesity is an independent, potent risk factor for ischemic stroke in all race-ethnic groups. It is a stronger risk factor than BMI and has a greater effect among younger persons. The goal of this experiment was to compare the global transcriptional profile, and its regulation, of white blood cells of healthy normals, patients with obesity and/or stroke in order to understand the transcriptional differences behind the different phenotypes. There were 6 subjects in each group. RNA was purified from white blood cells using a TRIZOL RNA Isolation Protocol (Life technologies) and 500 ng of starting material were used as input for the Illumina TotalPrep Amplification Kit. Next, the Array Studio (OmicSoft Corporation) was used to generate a list of genes that are expressed at a significantly higher/lower level using the two-way ANOVA in order to analyze the 2x2 factorial design.

Project description:Atrial fibrillation (AF) is a major risk factor for cardioembolic stroke. Anticoagulant drugs are effective in preventing AF-related stroke. However, the high frequency of anticoagulant-associated major bleeding is a major concern particularly when antiplatelet treatment is simultaneously administered. Here, microarray analysis in peripheral blood cells in eight patients with AF and stroke and eight AF subjects without stroke identified a stroke related gene expression pattern. HSPA1B, which encodes for heat-shock protein 70 kDa (Hsp70), was the most differentially expressed gene. This gene was downregulated in stroke subjects, a finding confirmed further in an independent AF cohort of 200 individuals. Hsp70 knock-out (KO) mice subjected to different thrombotic challenges developed thrombosis significantly earlier than their wild-type (WT) counterparts. In WT mice, Hsp70 inducers (TRC051384, or tubastatin A) delayed thrombus formation. Remarkably, Hsp70 inducers did not increase the bleeding risk even when aspirin was concomitantly administered. Hsp70 induction was associated with an increased vascular thrombomodulin expression, higher circulating levels of activated protein C (APC) upon thrombotic stimulus and increased protection against endothelial apoptosis. Thus, Hsp70 induction is a novel approach to delay thrombus formation with minimal bleeding risk, being especially promising in situations where there is a major bleeding hazard. Project description:Application of acute therapies such as thrombolysis for ischemic stroke (IS) is constrained because of diagnostic uncertainty and the dynamic nature of stroke biology. To investigate changes in blood proteins after stroke and as a result of thrombolysis treatment we performed label-free quantitative proteomics on serum samples using high resolution mass spectrometry and long HPLC gradient (5 hours) combined with a 50cm column to optimize the peptide separation. We identified (FDR: 1%) and quantified a total of 574 protein groups from a total of 92 samples from 30 patients. Ten patients were treated by thrombolysis as part of a randomised placebo controlled trial and up to five samples were collected from each individual at different time points after stroke. We identified 26 proteins differently expressed by treatment group (FDR: 5%) and significant changes of expression over time for 23 proteins (FDR: 10%). Molecules such as fibrinogen and CRP showed expression profiles with a high potential clinical utility in the acute stroke setting. Protein expression profiles vary acutely in the blood after stroke and have the potential to allow the construction of a stroke clock and to have an impact on IS treatment decision making.

Project description:In the present exploratory study, we aimed to characterize circulating inflam-matory protein profiles associated with glycemic status in individuals with severe obesity. Using proximity extension assay-based (PEA) proteomics, we first identified inflammatory proteins differentially expressed across normoglycemia, prediabetes, and T2DM and examined their distributions at the individual level. We then applied unsupervised topological data analysis to determine whether multidimensional in-flammatory protein patterns define distinct patient subgroups beyond conventional glycemic classification. By combining proteomic data, clinical variables, and network analysis, this study provides a hypothesis-generating view of inflammatory hetero-geneity in obesity-related dysglycemia.

Project description:Brain myeloid cells, including infiltrating monocyte-derived macrophages (MDMs) and resident microglia play critical roles in regulating debris clearance following ischemic brain injury, shaping neuroinflammation evolution and neurological outcome, yet the underlying mechanism remains elusive. In this study, using single-cell RNA sequencing and fate mapping, we identified serotonin receptor Htr2b+ MDMs as a pivotal subgroup involved in resolving neuroinflammation within the ischemic penumbra. In addition, we find significant accumulation of Htr2b+ MDMs with enhanced phagocytosis and inflammation-resolving gene signatures in the peri-infarct region. Mechanistically, Htr2b activation promotes nuclear translocation of Tfe3, a key lysosomal regulator, enhancing lysosomal biogenesis and supporting sustained phagocytic activity. Myeloid-specific Htr2b ablation impairs debris clearance, exacerbates neuroinflammation and ischemic injury, while pharmacological Htr2b agonism or Tfe3 activation restores the lysosomal competence of brain MDMs and improves post-stroke neurological outcome. In conclusion, we demonstrate a novel Htr2b+ MDM subset that integrates serotonin signaling to license their intrinsic phagocytosis function, alleviating ischemic brain injury.

Project description:This dataset consists of single-cell RNA sequencing (scRNA-seq) data derived from Peripheral Blood Mononuclear Cells (PBMCs) of two ischaemic stroke patients (P66 and P79). The study follows a longitudinal design to identify temporal changes in the systemic immune response, with samples collected at two timepoints: the acute in-patient stage (V1) and at 6-9 months follow-up (V3). Here, four samples (P66_V1, P66_V3, P79_V1, P79_V3) were multiplexed using the 10x Genomics CellPlex protocol and processed as a single pool. Approximately 33,000 cells were loaded onto a 10x Chromium Controller using the Chromium Next GEM Single Cell 5' Reagent Kit. This specific library represents the Gene Expression (GEX) portion of a multi-modal immune profiling experiment. The primary objective of this study was to assess the dynamics of the circulating immune compartment following ischaemic stroke.

Project description:Monocyte-derived liver macrophages are critical in the pathogenesis of metabolic dysfunction-associated steatohepatitis (MASH) and liver fibrosis, but their recruitment mechanisms remain unclear. Serotonin (5-hydroxytryptamine, 5HT) is a conserved monoamine synthesized by tryptophan hydroxylase (Tph) 1 in peripheral tissues and Tph2 in the brain. We show that in mice housed at thermoneutrality and fed a high-fat, high-fructose diet, inhibition of peripheral serotonin (pe5HT) through genetic deletion of Tph1 prevents MASH independently of reductions in body weight. Liver flow cytometry and single-nuclei sequencing showed reduced pro-inflammatory Ly6Chigh monocytes, monocyte-derived Kupffer cells (moKCs), and lipid-associated macrophages (LAMs) in Tph1 KO mice. Tph1 deletion also decreased circulating monocytes, specifically Ly6Chigh monocytes. A single 5HT injection increased Ly6Chigh monocytes, while Tph1 KO mice had reduced monocytes without affecting bone marrow monocytes. Mechanistically, serotonin inhibition increases liver AMPK activity, and this is important for reducing CCL2 and monocyte recruitment. These findings link two ancient energy sensors, 5HT and AMPK, with obesity-associated liver fibrosis.

Project description:CO2-induced vasoreactivity is an important mechanism to remove CO2 from the brain. Our previous work has shown that in brain endothelial cells Gαq and Gα11, that can replace each other and induce a common intracellular pathway, are required for normal vascular reactivity to CO2 stimulation. Additionally, we found different effects on vascular diameter dependent on the brain area. In the cortex, vessel dilate as response to CO2 stimulation, in the retrotrapezoid nucleus (RTN) of the brainstem, vessels constrict upon CO2 stimulation. To examine the potential basis for the opposing effects of CO2 on the vascular reactivity in brainstem compared to other brain areas, we prepared cortical and brainstem vessel fragments of control and Gαq/11beKO mice and performed microarrays to determine mRNA expression.
