DEK1 deletion effect on Physcomitrella patens transcriptome


ABSTRACT: The main phenotype observed in DEK1 deletion (deltadek1) in P. patens is an absence of gametophore (leafy shoot) development. But, bud initiation the first step in gametophore development occurs. This experiments aims to compare transcriptome of wild-type and deltadek1 strains before budding onset, in 6 day old culture, and after budding onset, in 14 days old cultures. At the first time point both culture are morphologically similar as after 14 days wild-type displays young gametophores and deltadek1 has only arrested buds.

DEFECTIVE KERNEL1 (DEK1) of higher plants plays an essential role in position-dependent signaling and consists of a large transmembrane domain (MEM) linked to a protease catalytic domain and a regulatory domain. Here, we show that the postulated sensory Loop of the MEM domain plays an important role in the developmental regulation of DEK1 activity in the moss Physcomitrella patens.
