MRNA profile of 5-aza-CR and 5-aza-CdR treated celllines

ABSTRACT: 5-azacytidine and 5-aza-2'-deoxycytidine are both demethylating agents but they have shown different clinical efficiency. Since 5-azacytidine is incorporated into RNA it might have a different impact on the expression profile than 5-aza-2'-deoxycytidine. This study is designed to investigate the immediate action of the DNMTi on day 2 and the late heritable effects on day 8 after start of treatment.

Background
The hypomethylating agent 5-Azacytidine (5-Aza-CR) is the first drug to prolong overall survival in patients with myelodysplastic syndrome (MDS). Surprisingly, the deoxyribonucleoside analog 5-Aza-2'deoxycytidine (5-Aza-CdR) did not have a similar effect on survival in a large clinical trial. Both drugs are thought to exert their effects after incorporation into DNA by covalent binding of DNA methyltransferase (DNMT). While 5-Aza-CdR is incorporated into only DNA, 5-Aza-CR is ...[more]
