Differential gene expression in mutants of Arabidopsis FAX1 compared to wild type

Project description:DNA microarray analysis (Affymetrix ATH1 GeneChip) on mutant lines of the gene At-FAX1 (At3g57280) in Arabidopsis thaliana was performed as described in Li et al. (2015). In total we analyzed the following comparisons: (A) flowers: fax1 knockout (n = 5) versus wild type (n = 5); (B) flowers: FAX1 over-expressors (n = 8, 4-times each line ox#2, ox#4) versus wild type (n = 5); (C) stems: fax1 knockout (n = 4) versus wild type (n = 4). Project description:Ghd7 regulates a range of functions in growth and development in response to environmental cues to maximize the reproductive success of the rice plant. We used microarrays to detail the regulated genes in OX-Ghd7HJ19 transgenic plants. To gain clues to downstream genes regulated by Ghd7, we performed a microarray analysis using Affimetrix rice gene chips. Young leaves in vegetative stage (35-days old) and developing panicles (0.1cm) from field-grown OX-Ghd7HJ19 transgenic and wild-type plants with two biological replicates were used to isolate RNA for chip analysis.

Expression data from sound treated Arabidopsis thaliana

Project description:Sound wave can cause various developmental and physiological changes in plant. It strongly suggests the existence of sophisticated molecular mechanisms for sound perception in plant. Hence gene expression study is important. We investigated the global gene expression in Arabidopsis upon five different single frequency treatment. Sound treated Arabidopsis was used for RNA extraction and hybridization on Affymetrix microarrays. Five different frequency (250, 500, 1000, 2000 and 3000 Hz) was applied at 80 db on 20 days old plant for one hour. Immediate after treatment (0 hour) rosette samples were harvested.

Total RNA from 2-week-old Arabidopsis thaliana (ecotype Columbia) seedlings grown on MS plates was isolated using RNeasy Plant Mini Kit from Qiagen. Differentially expressed genes between WT and AtC3H17-overexpressing transgenic plants

Project description:We characterized the biological and molecular functions of AtC3H17, a unique Arabidopsis gene encoding a non-tandem CCCH zinc finger protein, in plant development. To investigate the downstream regulatory mechanisms of AtC3H17, whole genome microarray expression profiling was carried out. The experiment was designed to identify differentially expressed genes between WT and AtC3H17-overexpressing transgenic plants (AtC3H17 OXs) grown for 14 days under SD growth conditions. Interestingly, the most up-regulated genes were 12S seed storage globulin genes, CRUCIFERIN A (CRA1) and CRUCIFERIN 3 (CRU3), and seed oil-body protein genes such as OLEOSIN 1 (OLEO1) and OLEO2 in AtC3H17 OXs compared with WT. We performed quantitative RT-PCR and confirmed that transcription levels of CRU3, OLEO1, OLEO2, and 2S seed storage albumin 1 (At2S1) were higher in AtC3H17 OX seedlings than in WT seedlings. Differentially expressed genes between WT and AtC3H17 OX at 14 days after germination under SD conditions (8-h-light and 16-h-dark cycle) were explored in whole genome level. Two AtC3H17 OX samples were compared to WT sample as a control.
