Transcriptional profiling by array of microRNAs of human peripheral blood mononuclear cells - PBMC - from control individuals for Type 2 diabetes mellitus patients

ABSTRACT: The microRNA oligo microarrays were used to determine expression profiles of peripheral blood mononuclear cells from control healthy individuals and compare to type 2 diabetes mellitus (T2DM) patients, aiming the identification of possible disease related MicroRNAs.

Project description:The microRNA oligo microarrays from Agilent were used to evaluate behavioral differences in osteoblast-like cells from calvaria and bone marrow of rats induced to osteoporosis by ovariectomy. The miRNAs data were normalized using the quantile and after statistical analysis by moderate t-test with p-value ≤ 0.05 and subsequent fold change ≥ 1.5, 84 miRNAs differentially expressed in calvaria cells were detected and 55 miRNAs in bone marrow cells.

The effect of the Autoimmune regulator (Aire) gene on microRNA transcriptional profile of 3.10 mTEC cell line

Project description:Medullary thymic epithelial cells (mTECs) contribute to self-tolerance through the ectopic expression of peripheral tissue antigens (PTAs) in the thymus. Autoimmune regulator (Aire) is to date the best characterized transcriptional regulator known to at least partially coordinate PTA expression in mTECs. Furthermore the expression of Aire-dependent and -independent genes is also modulated by microRNAs. Given the transcriptional regulation exerted by Aire in mTEC cells and the role of miRNAs in post-transcriptional control, we used a Mus musculus mTEC cell line (3.10) which constitutively express Aire in culture) to knockdown Aire gene by means of siRNA transfection and then observe the effect of Aire knockdown in the transcriptional profile of miRNAs by means of oligomicroarrays. The Agilent oligomicroarrays were used to determine the large scale the miRNA transcriptional profiles of control or Aire-knockdown 3.10 mTECs.

Transcription profiling of aortic samples from newborn and 6 week old C57/black6 wild type mice

Project description:Aortic samples ranging from the aortic valve to the diaphragm were harvested from 15 newborn and 15 six-week old C57/black6 wildtype mice. Frozen samples of 5 animals each were pulverized and directly dissolved in RNA pure (PeqLab) to get three samples for each group. Total RNA was isolated using phenol/chloroform extraction.<br><br>For gene-expression analysis, 500 ng total RNA of each RNA sample was labeled using the Agilent single-color Quick-Amp Labeling Kit and hybridized on Agilent Whole Mouse Genome Microarrays (4x44K). For<br><br>analysis of microRNA, 200 ng total RNA of each sample was labeled using the Agilent miRNA Labeling Reagent and Hyb Kit, and hybridized on Agilent Mouse miRNA Microarrays (8x15K) according to the manufacturer's instructions.
