Temporal Transcriptional Profiling in response to cMYC and KrasG12D Expression and Inactivation in In Vitro Cultured Primary Mammary Epithelial Cells of Adult Virgin Mice

ABSTRACT: Mammary glands from 8 week old, female, virgin GEMM MMTV-rtTA/TetO-MYC/TetO-KrasG12D mice were dissociated into single cells and seeded into matrigel matrix for in vitro culture. To determine the gene expression changes which occur upon oncogene induction 5 days after seeding doxycycline was added to the media for 5 days, and then removed. Samples were taken of the following populations: 5 days on, 5 days on/7days off and never induced.

Project description:Single-cell RNA sequencing analysis was performed on primary pancreatic ductal adenocarcinoma derrived cell line from PiKP (P48-Cre; R26-rtTa-IRES-EGFP; tetO-LSL-KrasG12D/+; Trp53L/L) murine tumors. In this study we establish that oncogenic Kras is the predominant driver of T cell paucity and myeloid infiltration in the pancreatic tumor microenvironment. Then, we use scRNA seq analysis of cultured PiKP cells with and without Kras* extinction to identify immune related genes involved in driving Kras* mediated immune supression in PDAC.

Effect of Misexpression of Sox9 on Gastric Differentiation

Project description:To determine if Sox9 was sufficient to drive mucous differentiation in the gastric epithelium, we bred mice carrying alleles for ROSA26rtTA.IRES.EGFP, TetO-Sox9, and Fgf20Cre.GFP. In these mice, cells expressing Cre will induce rtTA from the ROSA26rtTA.IRES.EGFP allele. To explore transcriptional changes following adult misexpression of Sox9, we isolated RNA from the corpus of Fgf20Cre.GFP/Cre.GFP; ROSA26rtTA.IRES.EGFP/rtTA.IRES.EGFP; TetO-Sox9 pup (misexpression animals) and ROSA26rtTA.IRES.EGFP/rtTA.IRES.EGFP; TetO-Sox9 pup (control) animals and performed RNA-sequencing.
