Project description:Auditory cued fear conditioning is a widely used behavioral paradigm to investigate associative learning and memory formation. During conditioning, an animal learns that a particular auditory stimulus predicts an aversive stimulus, a foot shock. When later confronted again with this conditional auditory stimulus rodents typically respond with a cessation of movement. The purpose of this study was to analyze the effects of auditory fear conditioning on gene expression in the auditory cortex in a number of different paradigms that control for the effect of several behavioral factors. The different paradigms were M-^SpairedM-^T were the sound and the shock are paired so that the mouse associates the tone with the noxious stimulus the shock, M-^SsoundM-^T were only the sound was presented, M-^SshockM-^T were only the shock was presented, M-^SunpairedM-^T were the sound and the shock were presented one minute apart so that the mouse cannot form an association between the sound and the shock, M-^ScontextM-^T were no sounds or shocks were presented but the mouse was in the behavior chamber and M-^SHCM-^T were the mouse was taken directly from the cage.

Project description:To identify genes involved in the developmental process of Atlantic salmon smoltification, gene expression was compared between smolt and parr in tissues involved in osmoregulation (gill), metabolism (liver), imprinting (olfactory rosettes) and neuroendocrine control (hypothalamus and pituitary). Tissue samples were harvested from laboratory-reared parr and smolts on the same date. Smolts were distinguished from parr by size and appearance; developmental status was confirmed by physiological assays. Eight biological replicates (16 fish) balanced for sex and for dye were used in the liver, gill, olfactory rosette, and hypothalamus comparisons. Four male parr were compared to four male smolts and four female parr were compared to four female smolts; smolts were labeled with Alexa Fluor 555 on four arrays and with Alexa Fluor 647 on four arrays. Six biological replicates (12 fish) were used for the pituitary comparison (two female and four male).

Project description:Regulation of milk synthesis and secretion is controlled mostly through local (intra-mammary) mechanisms. To gain insight into the molecular pathways comprising this response, an analysis of mammary gene expression was conducted in 12 lactating cows shifted from twice daily to once daily milking. Tissues were sampled by biopsy from adjacent mammary quarters of these animals during the two milking frequencies, allowing changes in gene expression to be assessed within each animal. Using bovine-specific, oligonucleotide arrays representing 21,495 unique transcripts, a range of differentially expressed genes were found as a result of less frequent milk removal. A total of 12 microarrays were used in this study. Each array was hybridized with RNA derived from a mammary sample taken at each biopsy time point, such that the twice-a-day milked sample and once-a-day milked sample were contained on the same slide for each animal (12 animals). Six of the microarrays represented animals previously shown to demonstrate a âlargeâ reduction in milk yield, whereas the other six arrays represented animals that previously displayed a âsmallâ reduction in milk yield when milked once per day. Half of each yield loss group (three animals) was represented by once-a-day milked/twice-daily-milked samples for Cy5/Cy3 labelling, with the other half labelled with the opposite dye configuration. This meant that six of 12 arrays were 'dye swapped', but with biological replicates as opposed to identical replicates.

Project description:Brucellosis is still a widespread zoonotic disease. Very little is known about the interaction between B. abortus and trophoblastic cells, which is essential for better understanding the pathogenesis of the Brucella-induced placentitis and abortion, a key event for transmission of the disease. The goal of this study was to evaluate the profile of gene expression by bovine trophoblastic cells during infection with B abortus. Explants of chorioallantoic membranes were inoculated with B. abortus strain 2308. Microarray analysis was performed at 4 h after infection, and expression of cytokines and chemokines by trophoblastic cells was assessed by real time RT-PCR at 6 and 12 h after inoculation. In addition, cytokine and chemokine expression was evaluated in placentomes from experimentally infected cows. Expression of pro-inflammatory genes by trophoblastic cells was suppressed at 4 h after inoculation, whereas a significant up-regulation of CXC chemokines, namely CXCL6 (GCP-2) and CXCL8 (IL-8), was observed at 12, but not at 6 h after inoculation. Placentomes of experimentally infected cows had a similar profile of chemokine expression, with upregulation of CXCL6 and CXCL8. Our data indicate that B. abortus modulates the innate immune response by trophoblastic cells, suppressing expression of pro-inflammatory mediators during the early stages of infection that is followed by a delayed and mild expression of pro-inflammatory chemokines, which is similar to the profile of chemokine expression in the placentomes of experimentally infected cows. This trophoblastic response is likely to contribute to the pathogenesis of B. abortus-induced placentitis. Keywords: trophoblast response to Brucella Total RNA used for microarray analysis was obtained from 6 placentas with 12 infected and 12 control explants from each placenta. At 4 h after inoculation, total RNA was isolated from the trophoblastic surface of the explants. Three RNA pools from two placentas each were generated prior to cDNA synthesis.
