Response of two clavata 1 mutants to Ralstonia solanacearum.

ABSTRACT: In this experiment, we compared transcriptome of 2 homozygous knock out mutants in the CLV1 gene (clv1-12 and clv1-13) to their wild type Ws-2 when challenged with the virulent Rd15 strain of Ralstonia solanacearum. The two mutants show a significant delay of symptom appearance. The experiment was conducted on roots and leaves of 4 week old plants harvested at the time of inoculation and when the first symptoms appeared on the susceptible wild type (4 days after inoculation).

Transcriptom analysis of the interaction Medicago truncatula – Ralstonia solanacearum

Project description:affy_ralstonia_medicago - Ralstonia solanacearum is the causal agent of the devastating bacterial wilt disease. Its infection process was studied with an in vitro inoculation procedure on intact roots of Medicago truncatula. The pathosystem involved susceptible A17 and resistant F83005.5 M truncatula lines infected with the pathogenic strain GMI1000. The mutant A17 line, Sickle, which showed a resistant phenotype was also part of the experiment. To identify host signaling pathway triggered by R. solanacearum infection with a focus on the involvment of ethylene, we used the Medicago Affymetrix array to monitore the expression profiles and the molecular process associated with initial symptoms development (12hpi) and colonization (72hpi). In order to maximize chances to observe differential gene expression, RNA samples were extracted from the root infection zone (root tips) -Three Medicago truncatula lines, A17, F83005.5 and sickle were inoculated with GMI1000 Ralstonai solanacearum strain (107 cfu/ml). RNA were extracted from root extremities (1 cm above the root tip) at time 0, 12h and 72h post inoculation. Ralstonia solanacearum effector-specific effect on Medicago truncatula trancriptome

Project description:affy_ralstonia_peeters_medicago - We have identified two essential virulence determinants (GALA7, a type III secretion effector and HpaP, a chaperone-like protein) of Ralstonia solanacearum for the infection and colonisation of the host plant Medicago truncatula. The scope of this project is to identify the GALA7 and HpaP-specific transcriptome alteration. For this purpose wild type and mutant infected root material (13h and 72h postinfection) will be analysed on M. truncatula affymetrix chips. Medicago truncatula (A17 line) are grown in vitro on Farheus medium (with Nitrogen source) plantlets are inoculated with water R. solanacearum wt, gala7 and hpap mutants, and root tips are collected at 13h and 72h postinoculation. Project description:Investigation of whole genome gene expression level changes in the bacterial wilt pathogen Ralstonia solanacearum, strain GMI1000 at 20°C and 28°C in culture and in planta. The tropical strain GMI1000 cannot wilt tomato plants at 20°C although it can cause full-blown disease at 28°C. A 16 array study using total RNA recovered from the following: 8 separate cultures of Ralstonia solanacearum strain GMI1000 grown in rich medium-CPG (4 grown at 20°C and 4 grown at 28°C) 8 separate in planta samples of Ralstonia solanacearum strain GMI1000 harvested from diseased tomato plants cv. BonnyBest (4 recovered from plants grown at 20°C and 4 from plants grown at 28°C) Each array (4-plex format) measures the expression level of 5061 genes from Ralstonia solanacearum strain GMI1000 with 2 to 6, 40-70 mer probes per gene, with two-fold technical redundancy. The arrays also contain probes for intergenic regions with no technical replicates.

Expression data from Ralstonia solanacearum-inoculated tomato stem

Project description:Bacterial wilt caused by Ralstonia solanacearum is a lethal, soil-borne disease of tomato. Control of the disease with chemicals and crop rotation is insufficient, because the pathogen is particularly well adapted for surviving in the soil and rhizosphere. Therefore, cultivar resistance is the most effective means for controlling bacterial wilt, but the molecular mechanisms of resistance responses remain unclear. We used microarrays to obtain the characteristics of the gene expression changes that are induced by R. solanacearum infection in resistant cultivar LS-89 and susceptible cultivar Ponderosa. Seedlings of LS-89 and Ponderosa at the five to six leaf-stage were inoculated on their stems just above the cotyledon by cutting to one-third the stem depth with a knife, adding a drop of bacterial suspension (1e+6 cfu/ml of R. solanacearum strain 8107S) or distilled water to the opening, and then clipping the wound site to avoid bending. Inoculated plants were grown in a growth chamber at 30ÂºC under 30,000 lux light intensity for 12 h a day. At 1dpi, stems were sampled by dissecting 5 mm long sections from 5 mm below the inoculation site. For each hybridization, RNA from 15 plants was used. Three biological replicates of microarray analysis were performed.

Gene expression analysis of Mulberry root after inoculation with Ralstonia solanacearum.

Project description:Bacterial wilt, caused by the soil-borne bacterium Ralstonia solanacearum, is a lethal disease of mulberry, but the molecular mechanisms of the host resistance responses to R. solanacearum remain unclear. In order to better understand molecular resistance mechanisms to R. solanacearum in mulberry, we set out to define the changes in gene expression of resistance and susceptible mulberry cultivars after inoculation with R. solanacearum. Susceptible cultivar YSD10, resistance cultivar KQ10 and YS283 were inoculation with R. solanacearum, mulberry root samples were collected at 1 dpi and non-treated control in all cultivars. Then we performed RNA-Seq analyses on all mulberry root samples using Illumina HiSeq 2000.
