Transcriptional profiling of DPB3-1 overexpressing rice plants under normal and heat stress conditions

ABSTRACT: Transcriptional profiles were compared between vector control and DPB3-1-overexpressing rice plants under control and heat stress conditions. Transcriptional changes of the vector control plants in response to heat stress were also studied.

Project description:BPMs are a family of E3 ubiquitin ligases. We identified BPM proteins as interactors of the Arabidopsis stress responsive transcription factor DREB2A. We found an increase in the accumulation level of the DREB2A protein under heat stress in transgenic Arabidopsis plants in which expression of six BPMs were suppressed by an amiRNA. In order to evaluate the impacts of the amiRNA on the gene expression under heat stress, transcriptomes were compared between these transgenic plants and vector control plants.

Transcription profiling of Arabidopsis thaliana hsc70-1/2/3 triple mutant under salt stress

Project description:HSC70 is the cytosolic isoform of plant HSP70. We have found that HSC70 family proteins bind to the heat shock transcription factor A1s (HsfA1s), which are the master regulators of the heat shock response in plants, and suppress their activity. We additionally found that the triple knock out of HSC70s alters responses of Arabidopsis plants to salt stress. To investigate the role of the HSC70s in salt stress responses, we evaluated the effects of the triple knock out on the transcriptome under salt stress.

Project description:In this series of experiments, we wanted to study the transcriptional responses of plants to different levels of water limitation. For mild drought stress, we controlled water potential (a scientific concept for dryness of soil) by using an automated watering system. This system adds water to soil based on the pF values reported by a pF sensor in soil. pF is a classical and widely used index of water potential that was first defined by Schofield (1935). For severe stress conditions, we withheld watering or even dried plants on a lab bench. Here, the transcriptional profiles were compared between well-watered and Sds-treated rice seedling shoot basal region. Sds (Severe dehydration stress in soil containers): We designated Sds as a term that means a severe drought stress causing dehydration of plants in our experiments (The plants were dehydrated at the sampling point). The drought stress was achieved by withholding water from plants grown in soil containers.

Comparative analysis of heat-induced genes between thermotolerant and thermosensitive tomato cultivars (50°C 1 h)

Project description:Gene expression in response to severe heat stress (50°C 1 h) was compared between thermotolerant and thermosensitive tomato cultivars. The thermosensitive cultivar showed severe impairment of heat stress response at this condition, while the thermotolerant cultivar were less affected. The results of this experiment were used to select expression marker genes that reflect the cellular heat shock response and correlate with thermotolerance.
