Transforming growth factor-B1 drives differentiation of the embryonic jejunum

ABSTRACT: Transcription profiling by microarray was performed to pinpoint the roles of transforming growth factor B1 (TGFB1) in embryonic differentiation of the small intestine. An organ culture system in which mouse jejunal rudiments were placed on semipermeable platforms and fed with chemically defined serum-free media, supplemented with two concentrations of TGFB1.

Project description:Oesophageal adenocarcinoma (OAC) is one of the ten most prevalent forms of cancer which is showing a rapid increase in incidence and yet exhibits poor survival rates. Compared to many other common cancers, the molecular changes that occur in this disease are relatively poorly understood although genomic sequencing studies have identified several genes encoding chromatin remodeling enzymes that are frequently mutated in OAC. This finding is consistent with the knowledge that one important change that occurs in cancer cells is a reprogramming of the chromatin environment which leads to subsequent changes in their transcriptional profile.

Circadian clock study of mice tail tendons

Project description:Tendons are prominent members of the family of fibrous connective tissues (FCTs), which collectively are the most abundant tissues in vertebrates and have crucial roles in transmitting mechanical force and linking organs. Tendon diseases are among the most common arthropathy disorders; thus knowledge of tendon gene regulation is essential for a complete understanding of FCT biology. Here we show autonomous circadian rhythms in mouse tendon and primary human tenocytes, controlled by an intrinsic molecular circadian clock. Time-series microarrays identified the first circadian transcriptome of murine tendon, revealing that 4.6% of the transcripts (745 genes) are expressed in a circadian manner.

Project description:This experiment is part of the FunGenES project (FunGenES - Functional Genomics in Embryonic Stem Cells partially funded by the 6th Framework Programme of the European Union, http://www.fungenes.org). The experiment was conducted at the Inst. National de la SantÃ© et de la Recherche MÃ©dicale - INS (INSERM U371, Bron, France) The goal of the experiment is to identify genes regulated by STAT3 and NANOG, which are known inhibitors of mouse ES cell differentiation.(I) E14-Cre-CTL cells deprived of LIF for 24 hours (n = 5); (II) E14-Cre-Tax cells as in (I) and restimulated with 100 nM tamoxifen for 2 hours (n = 5); (III) E14-STATA3-CTL cells deprived of LIF and 4âOHT for 24 hours (n = 4); (IV) E14-STAT3-LIF cells treated as in (III) and re-stimulated with 10,000 U/ml LIF for 1 hour to reactivate endogenous STAT3 (n = 5); (V) E14-STAT3-Tax cells as in (III) and restimulated with 100 nM 4âOHT for 2 hours to activate STAT3-ERT2 (n = 6);

Ectopic Hedgehog signalling causes cleft palate and defective osteogenesis

Project description:In this study, we have investigated the molecular basis of Shh signalling during development of the secondary palate and how CNCC patterning and fate is influenced by the Shh signalling network. Using a gain-of-function mouse model to activate Smoothened (R26SmoM2) signalling in the palatal mesenchyme (Osr2-IresCre), we demonstrate ectopic Hh-Smo signalling results in fully penetrant cleft palate, disrupted oral-nasal patterning and defective palatine bone formation. We show that a series of Fox transcription factors, including the novel direct target Foxl1, function downstream of Hh signalling in the secondary palate. Furthermore, we demonstrate that Wnt/BMP antagonists, in particular Sostdc1, are positively regulated by Hh signalling, concomitant with down-regulation of key regulators of osteogenesis and BMP signalling effectors. Microarray analysis was performed on excised palatal shelves from Osr2-IresCre+/- (wild-type) and Osr2-IresCre;Smo+/M2 (mutant) embryos at embryonic day (E)13.5. Osr2-IresCre (PMID:17941042) and R26SmoM2 (PMID:15107405) mice have been described previously.
