ABSTRACT: Experiment Description RNA sequencing was performed on Candida albicans wild type cells (SC5314) grown to exponential phase on YNB Lactate, YNB Glucose, or YNB Glucose plus Lactate, and compared to exponential Candida albicans crz1 cells grown on YNB Glucose or YNB Glucose plus Lactate. Three independent experiments were performed.

Robust extracellular pH modulation by Candida albicans during growth in carboxylic acids

Project description:Purpose: Candida albicans avidly uses multiple carbon sources that are less preferred by related species. This project compared transcriptional profiles of cells growing in media containing these nutrients relative to glucose Methods: C. albicans strain SC5314 was incubated in minimal YNB media containing 2% glucose, casamino acids, glutamate or a-ketoglutarate for five hours. RNA was prepared and subjected to deep sequencing. Results: Substantial overlap was seen in the transcriptional changes in all alternative carbon sources relative to the glucose control. Source-specific changes reflected the unique metabolism of each nutrient. Conclusions: C. albicans is well-adapted to use a range of nutrients that can be found in the mammalian host and has a common transcriptional response to limiting carbon environments.

Lactate- and hypoxia-induced beta-glucan masking in responding and non-responding Candida albicans clinical isolates

Project description:RNA sequencing was performed on Candida albicans clinical isolates that display normal (isolates: 3560, 3605, 3609, 4108, 4259) or aberrant (isolates: 3534, 3544, 3621, 3636, 4036) beta-glucan masking in response to lactate and hypoxia. Each clinical isolate was grown to exponential phase in GYNB under normoxic conditions and then exposed for 5 h to: (a) 1% lactate; (b) 0% lactate control; (c) hypoxia; or (d) normoxic control. Three independent experiments were performed for each clinical isolate.

This extracellular armour is critical for fungal cell homeostasis and survival. Yet essential cell wall moieties, such as β-1,3-glucan, are recognised as pathogen-associated molecular patterns (PAMPs) that activate immune-mediated clearance mechanisms. We have reported that the opportunistic human fungal pathogen, Candida albicans, masks β-1,3-glucan following exposure to lactate, hypoxia or iron depletion. However, the precise mechanism(s) by which C. albicans masks β-1,3-glucan have remained obscure. Here, we performed proteomic analysis of supernatants harvested from C. albicans cells grown in hypoxia or lactate compared to glucose-grown controls to identify mechanisms driving β-1,3-glucan masking.

Expression analysis of Candida albicans WO-1 grown in glucose or xylose

Project description:Investigation of whole genome gene expression level changes in Candida albicans WO-1 grown aerobically in xylose, compared to the same strain grown aerobically in glucose. A six array study using total RNA recovered from three separate cultures of Candida albicans WO-1 grown in glucose and three separate cultures of Candida albicans WO-1 grown in xylose. Each array measures the expression level of 373,067 probes (average probe length 55.2 +/- 4.0 nt) tiled across the Candida albicans WO-1 genome with a median spacing distance of 31 nt. During data processing, probes are filtered to include only those probes corresponding to annotated protein-coding genes.

Project description:To examine the role of a glycosylphosphatidylinositol-linked aspartyl protease, CgYps1, in the regulation of pH homeostasis in Candida glabrata, transcriptional profiling analysis was carried out on wild-type and Cgyps1∆ cells grown in YNB medium (pH 5.5) and in YNB medium adjusted to pH 2.0. Genes involved in carbohydrate and amino acid metabolism, protein folding and stress response pathways were found to be differentially regulated in response to acidic environment in both the strains.
