Transcript profiling of the biotrophic interaction between Theobroma cacao and the fungal pathogen Moniliophthora perniciosa

ABSTRACT: RNA-seq to simultaneously inspect the transcriptomes of cacao and M. perniciosa during their biotrophic interaction. Five biological replicates of each condition (healthy seedlings and seedlings at 30 d after infection) were harvested for transcriptome sequencing using RNA-seq. For this experiment, the entire green broom structure was collected, including cacao leaves and shoots. Samples in SRA were assigned the same sample accession (SRS396688, SRS396689). This is incorrect as there are different samples, hence “Source Name” was replaced with new values. Comment[ENA_SAMPLE] contains the original SRA sample accessions.

INSTRUMENT(S): Illumina HiSeq 2000

Witches' broom disease (WBD), caused by the hemibiotrophic fungus Moniliophthora perniciosa, is one of the most devastating diseases of Theobroma cacao, the chocolate tree. In contrast to other hemibiotrophic interactions, the WBD biotrophic stage lasts for months and is responsible for the most distinctive symptoms of the disease, which comprise drastic morphological changes in the infected shoots. Here, we used the dual RNA-seq approach to simultaneously assess the transcriptomes of cacao and  ...[more]
