Tissue compartmentalization and distinct stem cell behavior underlie epidermal homeostasis

Project description:Long-term tissue homeostasis is governed by balanced contribution from adult stem cells. We recently identified Lrig1 as a marker of stem cells in mouse epidermis. Here, we show that Lrig1 expressing cells are molecularly and functionally distinct from previously characterized epidermal stem cell populations. During steady state homeostasis, Lrig1 expressing cells are responsible for the maintenance of the uppermost compartment of the pilosebaceous unit known as the infundibulum. Lineage tracing demonstrates that the epidermis is divided into discrete compartments during homeostasis, each maintained by its own resident stem cells. Compartment boundaries are rapidly broken when stem cell progeny are recruited to sites of injury, where they subsequently change behavior according to the environment. Oncogene activation in Lrig1 expressing cells alters proliferation but auxiliary stimuli are required for rapid tumor growth. Our data demonstrate that stem cell niches are compartmentalized according to altering requirements for tissue replenishment.

Project description:RNA-seq of embryonic mammary progenitor cell lines. Embryonic mammary progenitor cell (eMPC) clones were derived from E12.5-stage mammary organs. After microdissection, mammary primordial 3 was cultured on thick basement membrane extract for 4 weeks. After enzymatic dissociation eMPC swere plated and expanded in 2D culture to obtain a pool of eMPCs. eMPC cells were subject to single cell sorting using FACS. Clones were expanded as single-cell derived clones (eMPC1, 2, etc.) to create the eighteen single cell-derived clones described in this study.

Microarray expression profiling of the Igf2 pathway dependency of Trp53 developmental and tumour phenotypes in mice

Project description:We tested whether loss of p53 function leads to insulin-like growth factor 2 (IGF2) pathway dependency in vivo in genetically defined mouse models. Unexpectedly we found lethality, due to post-natal lung haemorrhage, occurred in Igf2 paternal null allele female mice (Igf2-p) but only if derived from double heterozygote fathers (Igf2-p, p53+/-). Consequently we investigated the effects of paternal genotype (wild-type, Igf2-p and Igf2-p, p53+/-) on gene expression. Microarray gene expression profiling of whole mouse embryos (embryonic day 9.5) revealed that lethality was associated with a specific gene signature. Since double heterozygote female offspring (Igf2-p, p53+/-) of Igf2-p, p53+/- fathers did not have the lethality phenotype we identified gene expression changes associated with this 'rescue'. Project description:Transcriptomic profiling of gene expression in EA 2018 relative to that of ATCC824 revealed several key genes related to solvent formation. For example, spo0A and adhEII have higher expression level, and most of the acid formation related genes have lower expression level in EA 2018. Cells for RNA isolation were grown on P2 medium and collected at 9 h, 13 h, 17 h, 21 h, 24 h and 30 h by centrifugation at 4M-BM-0C and 4500 x g for 10 min. Total RNA was extracted and purified by using Trizol (Invitrogen, Carlsbad, CA, USA) and RNeasy cleanup kit (Qiagen, Inc., Valencia, CA, USA) according to the manufacturerM-bM-^@M-^Ys protocol. The total RNA yield was quantified by spectrophotometric analysis (NanoDrop Technology, Cambridge, UK) and the quality was verified by gel electrophoresis. Agilent oligonucleotide microarrays technology was used for monochromic analysis, in which probes (size: 60bp; three replicates for each ORF) from the two groups were labeled by incorporation of cyanine 3 (Cy3) (Agilent Technologies, Palo Alto, CA, USA). Average linkage hierarchical clustering was performed using Cluster 3.0, and gene clusters were visualized in Treeview
