RNA-Seq of murine lung endothelial cells stimulated with IFN-gamma and dexamethasone

ABSTRACT: Glucocorticoids are potent anti-inflammatory drugs prescribed in various diseases such as asthma, rheumatoid arthritis and multiple sclerosis. The effects of glucocorticoids have extensively been studied in immune cells. However, the regulation of inflammation by glucocorticoids in endothelial cells remains poorly understood. In this study, we stimulated murine lung endothelial cell line with dexamethasone (100 nM) and IFN-gamma (20 ng/mL) for 24 hours and extracted RNA for RNA-seq.

Project description:Patients with the diffuse avascular form of Systemic Sclerosis (SSc) and healthy controls (N) were used as sources of microvascular endothelial cells (MVEC). MVEC were obtained from biopsies of involved skin of the hands in 6 SSc patients and from 6 healthy patients undergoing surgery for traumatic events at the hands. The controls were matched by age and sex to the SSc cases. All patients fulfilled the American College of Rheumatology criteria for SSc, as well as the criteria for the diffuse form of the disease.

Project description:To investigate the specific roles of SIRT1 in the development of hepatocellular carcinoma, we employed large-scale gene expression analysis to identify the molecular signature that may affect enabling characteristics of cancer cells. Differentially expressed genes were analyzed on the SNU-182 cells transfected with SIRT1 siRNA and recapitulated molecular signatures that related to hallmarks of cancer. Thus, in this study we want to analyze the transcriptomic alterations in VAT during active and cured CS and correlate these persistent gene expression changes with histone modifications induced by GCs.

Comparative Gene Expression Profiling of HUVEC and Ocular Vascular Endothelial Cells

Project description:To compare the gene expression profiles of unpassaged, proliferating HUVEC and human iris, retinal and choroidal microvascular endothelial cells. Gene expression profiling revealed significant differences between HUVEC and ocular microvascular endothelial cells suggesting that HUVE cells may not be a suitable surrogate when studying pathophysiological mechanisms of ocular disorders. There were significant differences in the gene expression of important cell signalling pathways in human retinal and choroidal ECs. These differences may be important in the mechanisms and treatment of choroidal and retinal neovascularisation. 12 arrays are included. Endothelial cells were derived from 4 tissues: iris, retina, choroid and human umbilical vein. RNA extracts from cells were hybridised to Affymetrix HGU133plus2 arrays in triplicate.
