Irisin is a Pro-Myogenic Factor that Induces Skeletal Muscle Hypertrophy and Rescues Denervation-Induced Atrophy

ABSTRACT: C2C12 myoblasts were seeded at a density of 25,000 cells/cm2 in a 10cm cell culture dish and differentiated with low serum differentiation medium at 37 °C, 5% CO2 for 72h and further treated with either DB or recombinant Irisin protein (1000ng/ml) for 6h, 12h, 24h and 48h. Myotubes were resuspended in 2ml of TRIzol ® for each 10cm dish and RNA isolation was performed according to the manufacturer s protocol. Purified isolated RNA was then subjected to Illumina bead array sequencing and data analysis, as per in-house protocols (Sciencewerke, Singapore). Only genes that were upregulated or downregulated by 1.5-fold were considered significant. For genes that had multiple probes (with different accession number), only one probe with the highest upregulation or downregulation was taken into consideration.

Project description:Analysis of platelet-derived growth factor (PDGF)-stimulated fibroblasts. Results provide insight into novel pro-tumorigenic factors induced by PDGF-activation of fibroblasts. 1.2 x 10e6 BJ-hTERT fibroblasts were cultured in 10cm dish with Dulbecco's Modified Eagle Medium (DMEM; Gibco Life Technologies, Gergy-Pontoise, France) containing 1% heat-inactivated fetal calf serum (FCS), 2 mM L-glutamine, penicillin (100 units/ml) and streptomycin (100 ng/ml) at 37ÂºC in a 5% CO2 humidified atmosphere during 24h. Cultures were stimulated with or without 20ng/mL PDGF-BB (Peprotech, New Jersey, USA) for another 24h prior to harvest. Unstimulated fibroblasts were used as control samples. Cultures of both unstimulated and stimulated fibroblasts were done in triplicate.

qXL-MS of insulin resistance in C2C12 myotubes

Project description:C2C12 myotubes were made insulin resistant with 0.5 mM palmitic acid for 24 h along with non-treated controls and then each group stimulated with or without 100 nM insulin for 30 min (n=4 each). Cells were then cross-linked with 2 mM of tPhoX for a further 30 min in the presence of insulin and the reaction quenched as previously described (Jiang et al., 2022). To increase the depth of analysis, each sample was subjected to a crude biochemical-based separation into four fractions (Wong et al., 2025) followed by digestion with trypsin/LysC. The 16 samples from each fraction were labelled with 16-plex Tandem Mass Tags (TMT) and pooled, followed by the depletion of phosphopeptides and enrichment of XL-peptides using immobilised metal-ion affinity chromatography (IMAC). Each of the four biochemical fractions underwent peptide-level size-exclusion chromatography (SEC) to partially fractionate larger XL-peptides from mono-/loop-linked peptides in 2-3 fractions which were further separated by high pH reversed-phase chromatography into 12 fractions and analysed by LC-MS/MS using data-dependent acquisition (DDA) resulting in the analysis of 132 individual runs. Data were analysed with pLink2 and searched against a skeletal muscle-specific mouse proteome database to limit the search space with filtering to 1% FDR followed by reporter-ion quantification and statistical analysis

PCP-MS analysis of insulin resistance in C2C12 myotubes with or without acute insulin stimulation

Project description:To study dynamic remodelling of protein complexes, C2C12 myotubes were treated with 0.5 mM palmitic acid for 24 h to induce insulin resistance (IR) and then stimulated with or without acute 100 nM insulin for 30 min followed by PCP-MS(n=3 each. Samples were lysed in non-denaturing buffer, separated by BN-PAGE, and each lane cut into 30 x 1.5mm fractions. Each fraction underwent in-gel digestion and was analysed with liquid chromatography – tandem mass spectrometry (LC-MS/MS) using data-independent acquisition (DIA) resulting in the analysis of 360 individual runs.

Genome-wide survey of YY1 targets in C2C12 cells by High-throughput sequencing

Project description:By applying ChIP-seq, we generated genome-wide maps of YY1 in skeletal myoblasts and myotubes. We found that YY1 binds to 1820 confident target with a large portion residing in the intergenic regions. In addition, YY1 was found to activate many loci, and there is no significant overlap between YY1 and Ezh2 targets, suggensting a Ezh2-independent manner. Further detailed study revealed that YY1 can regulate some lincRNAs which are fucntional in skeletal myogenesis. Cancer-cell-secreted miR-122 suppresses O-GlcNAcylation to promote muscle proteolysis

Project description:Decline in skeletal muscle mass and lower muscular strength are prognostic factors in advanced human cancers. We found that breast cancer suppressed O-linked N-acetylglucosamine (O-GlcNAc) protein modification in skeletal muscle through extracellular-vesicle-encapsulated miR-122 that targeted O-GlcNAc transferase (OGT). Specifically, O-GlcNAcylation of ryanodine receptor 1 (RYR1) Ca2+ release channel competed with NEK10-mediated phosphorylation and increased K48-linked ubiquitination and proteasomal degradation, whereas O-GlcNAcylation of sarco/endoplasmic reticulum Ca2+-ATPase (SERCA3) increased K63-linked ubiquitination and elevated protein level. Muscular protein O-GlcNAcylation was regulated by hypoxia and lactate through HIF1A-dependent OGT promoter activation, and elevated after exercise. Suppressed O-GlcNAcylation in the setting of cancer, especially through increased RYR1 protein, led to Ca2+ leak and calpain protease activation to trigger myofibrillar protein cleavage. This was associated with reduced skeletal muscle mass and contractility in tumor-bearing mice. Our findings link O-GlcNAcylation to muscular protein homeostasis and contractility, and reveal a mechanism of cancer-associated muscle dysregulation.
